安宮牛黃丸HPLC指紋圖譜及其多成分化學(xué)模式分析

【摘要】:目的建立安宮牛黃丸(ANP)HPLC指紋圖譜,測定其中6種有效成分的量并進(jìn)行化學(xué)模式分析。方法采用HPLC法,以乙腈-0.1%磷酸水溶液為流動(dòng)相進(jìn)行梯度洗脫,檢測波長為230 nm,對(duì)4個(gè)不同廠家生產(chǎn)的12批ANP樣品進(jìn)行測定,并采用中藥色譜指紋圖譜相似度評(píng)價(jià)軟件進(jìn)行相似度分析。對(duì)主要有效成分梔子苷、京尼平苷酸、鹽酸小檗堿、黃芩苷、漢黃芩苷、漢黃芩素進(jìn)行了定量測定。并采用化學(xué)模式識(shí)別的方法對(duì)不同廠家產(chǎn)品進(jìn)行分析。結(jié)果 4個(gè)不同廠家生產(chǎn)的12批ANP的指紋圖譜相似度均在0.9以上。對(duì)6種有效成分定量測定結(jié)果發(fā)現(xiàn)不同廠家產(chǎn)品仍存在一定差異,尤其以黃芩所含成分差別較大,說明ANP中黃芩原料藥材差異較大,其中C廠家產(chǎn)品中黃芩苷、漢黃芩苷及漢黃芩素的量較其他廠家更為穩(wěn)定,并且其量普遍高于其他廠家。通過主成分分析中的F綜合評(píng)分,進(jìn)一步評(píng)價(jià)不同廠家樣品質(zhì)量的優(yōu)劣。結(jié)果發(fā)現(xiàn),C廠家產(chǎn)品在綜合排名中排名較靠前,其結(jié)果與其定量測定結(jié)果基本吻合。結(jié)論指紋圖譜結(jié)合有效成分的化學(xué)模式分析能更全面地反映ANP的質(zhì)量和優(yōu)劣,所建立的方法對(duì)進(jìn)一步合理控制ANP的質(zhì)量提供了實(shí)驗(yàn)依據(jù)。
[Abstract]:Objective to establish the (ANP) HPLC fingerprint of Angong Niuhuang Pill, determine the contents of 6 active components and carry out chemical model analysis. Methods gradient elution was carried out with acetonitrile-0.1% phosphoric acid aqueous solution as mobile phase by HPLC method. The detection wavelength was 230nm, for the determination of 12 batches of ANP samples produced by four different manufacturers. The similarity analysis was carried out by the similarity evaluation software of chromatographic fingerprint of traditional Chinese medicine. The main effective components of geniposide, berberine hydrochloride, baicalin and baicalin were determined quantitatively. The chemical pattern recognition method was used to analyze the products of different manufacturers. Results the similarity of 12 batches of ANP fingerprints from 4 different factories was above 0.9. The results of quantitative determination of six kinds of active components showed that there were still some differences among the products from different manufacturers, especially the components contained in Scutellaria baicalensis, which indicated that the raw materials of Scutellaria baicalensis in ANP were different greatly, among which baicalin was found in the products of factory C. Baicalin and baicalin were more stable than other manufacturers, and generally higher than other manufacturers. The F score of principal component analysis was used to evaluate the quality of samples from different factories. The results showed that the products of C manufacturers ranked higher in the overall ranking, and the results were basically consistent with the quantitative results. Conclusion fingerprint analysis combined with chemical model analysis of active components can reflect the quality and quality of ANP more comprehensively. The established method provides experimental basis for further rational control of ANP quality. [WT5HZ] conclusion\?
