紅外光譜法研究古代彩繪蛋白膠料光老化的二級結(jié)構變化

【摘要】:動物膠和蛋清是古代重要彩繪文物蛋白膠結(jié)材料,對其光老化蛋白質(zhì)結(jié)構變化的研究與探索彩繪文物病變機理和選擇科學保護材料、修復方法密切相關。以維也納藝術歷史博物館的動物膠(骨膠、兔皮膠、鱘魚膠)和蛋清為研究對象,利用紅外光譜技術,結(jié)合酰胺Ⅰ帶去卷積和高斯擬合法研究光老化過程中蛋白質(zhì)二級結(jié)構的改變。結(jié)果表明,四種膠光老化后仍然具備蛋白質(zhì)紅外光譜的典型吸收特征。但紅外吸收峰出現(xiàn)了不同程度的位移,其中蛋清糖苷鍵明顯藍移。酰胺Ⅰ帶去卷積和高斯擬合結(jié)果顯示,蛋白膠的二級結(jié)構占比發(fā)生了改變。其中,骨膠α-螺旋結(jié)構占比降低22.24%,逐漸解螺旋,無規(guī)則卷曲結(jié)構占比明顯增高(9.76%);兔皮膠和鱘魚膠α-螺旋結(jié)構占比分別降低5.31%,4.15%,前者無規(guī)則卷曲增幅最低(6.96%),后者反而減少,推測兩者的光老化穩(wěn)定性高。蛋清α-螺旋占比降低程度最高(44.45%),無規(guī)則卷曲占比增幅極大(27.49%),光老化穩(wěn)定性最差。證實利用紅外光譜技術,結(jié)合酰胺Ⅰ帶去卷積和高斯擬合法研究文物蛋白膠料二級結(jié)構變化的方法是可行的,但其轉(zhuǎn)化機理還需后期進一步探討。
