五味消毒飲口服液HPLC指紋圖譜的建立及6種指標性成分定量測定

【摘要】:目的建立五味消毒飲口服液(WXOL)的HPLC法特征指紋圖譜,并同時測定綠原酸、木犀草苷、木犀草素、蒙花苷、咖啡酸、秦皮乙素6種主要指標成分的量。方法采用Agilent 1260高效液相色譜儀進行色譜分析,色譜柱為Agilent TC-C18柱(250 mm×4.6 mm,5μm);流動相為0.5%醋酸水溶液-甲醇,梯度洗脫:0~10 min,10%~32%甲醇;10~20 min,32%甲醇;20~25 min,32%~46%甲醇;25~31 min,46%~48%甲醇;31~41 min,48%~80%甲醇;體積流量為1 m L/min;檢測波長為320 nm;柱溫為30℃;采用國家藥典委員會出版的《中藥色譜指紋圖譜相似度評價系統》(2012年版)對10批WXOL化學成分指紋圖譜進行相似度評價,并采用多成分同時測定法對6種指標性成分進行定量測定。結果 10批WXOL HPLC特征指紋圖譜相似度均大于0.98,共17個共有峰,各峰分離度較好。通過對照品比對確定了其中綠原酸(5號峰)、秦皮乙素(7號峰)、咖啡酸(8號峰)、木犀草苷(12號峰)、蒙花苷(16號峰)、木犀草素(17號峰)6個成分,10批WXOL中綠原酸量為54.038 3~105.551 1μg/m L,秦皮乙素量為4.122 1~31.359 9μg/m L,咖啡酸量為2.413 0~4.420 7μg/m L,木犀草苷量為4.042 8~11.312 8μg/m L,蒙花苷量為3.866 3~46.271 9μg/m L,木犀草素量為0.990 8~2.126 8μg/m L。不同批次間各指標性成分量變化較小,樣品質量較穩(wěn)定。結論所建立的HPLC特征指紋圖譜結合多指標成分定量測定的方法,穩(wěn)定性、重復性較好,有利于保證WXOL質量均一,對其質量控制具有參考價值。
