HPLC測定半枝蓮中野黃芩苷、黃芩素、木犀草素和芹菜素的含量

【摘要】:建立半枝蓮藥材中野黃芩苷、黃芩素、木犀草素和芹菜素含量同時測定的HPLC方法。色譜條件為DiamonsilC18柱(250 mm×4.6 mm,5μm)色譜柱分離,檢測波長為335 nm,流動相為乙腈(A)-1%冰醋酸水溶液(B)進行梯度洗脫,流速為1.0 m L/min,柱溫為30℃。野黃芩苷、黃芩素、木犀草素和芹菜素濃度分別在2.0~200、0.5~50、0.5~50、0.5~50μg/m L范圍內與峰面積積分值線性關系良好。在精密度實驗、穩(wěn)定性實驗、重復性實驗中,野黃芩苷、黃芩素、木犀草素和芹菜素峰面積的相對標準偏差(RSD)2.0%;野黃芩苷、黃芩素、木犀草素和芹菜素的加樣回收率范圍及平均值和RSD分別為97.73%~101.7%、99.72%、1.36%(n=9),97.78%~102.8%、99.53%、1.99%(n=9),97.40%~102.4%、99.39%、1.84%(n=9)和97.80%~102.0%、99.24%、1.49%(n=9)。所測12份不同產地半枝蓮藥材中,野黃芩苷、黃芩素、木犀草素和芹菜素的含量范圍分別為2.41~7.08、0.26~0.56、0.31~0.90、0.28~0.95 mg/g。本方法具有良好的專屬性和重現性,加樣回收率實驗符合要求,能準確、穩(wěn)定地對半枝蓮藥材中野黃芩苷、黃芩素、木犀草素和芹菜素含量進行同時定量測定。
