脂質(zhì)體平衡透析與液質(zhì)聯(lián)用技術(shù)篩選脊痛寧片物質(zhì)基礎(chǔ)研究

【摘要】:目的研究脊痛寧片有效成分與模擬生物膜的相互作用,預(yù)測(cè)藥物在體內(nèi)的吸收情況。方法將脂質(zhì)體作為模擬生物膜,采用脂質(zhì)體平衡透析與液質(zhì)聯(lián)用技術(shù)篩選脊痛寧片中與脂質(zhì)體有相互作用的成分,并考察脂質(zhì)體質(zhì)量濃度和緩沖液p H值對(duì)脊痛寧片與脂質(zhì)體互相作用的影響。結(jié)果脊痛寧片中共有11個(gè)成分與脂質(zhì)體作用明顯,包括4個(gè)黃酮類成分,2個(gè)萜類成分,4個(gè)香豆素類成分,1個(gè)苯丙酸類成分。脂質(zhì)體的質(zhì)量濃度和緩沖液p H值都對(duì)脊痛寧片與脂質(zhì)體互相作用有一定的影響。結(jié)論脂質(zhì)體平衡透析與液質(zhì)聯(lián)用可預(yù)測(cè)脊痛寧片潛在的活性成分在體內(nèi)的吸收情況,為其藥效物質(zhì)基礎(chǔ)的進(jìn)一步研究提供依據(jù)。
[Abstract]:Objective to study the interaction between the active components of Jitongning tablets and mimic biofilm and to predict the drug absorption in vivo. Methods Liposome was used as a mimic biofilm and liposome equilibrium dialysis and liquid-mass spectrometry were used to screen the components of Jitongning tablet that interact with liposome. The effects of liposome concentration and buffer pH on the interaction between Jitongning tablets and liposomes were investigated. Results there were 11 components in Jitongning tablets, including 4 flavonoids, 2 terpenoids, 4 coumarins and 1 phenylalanic acid. The mass concentration of liposome and pH value of buffer had some influence on the interaction between Jitongning tablets and liposomes. Conclusion Liposome equilibrium dialysis combined with liquid-mass spectrometry can predict the absorption of potential active components in Jiitongning tablets in vivo, which provides a basis for further study on the basis of its pharmacodynamic substance.
