【摘要】：單向兩次薄層層析展開可以顯著提高對熱凝膠β-1,3-葡寡糖的分析效果,以Silica gel60 F254硅膠板為固定相,V(乙酸)∶V(乙酸乙酯)∶V(水)=2∶2.4∶1.4為展開劑,展距20 cm時,單向兩次展開可以較好地分離聚合度為2~13的熱凝膠β-1,3-葡寡糖。此外還建立了Image J軟件圖像分析定量熱凝膠β-1,3-葡寡糖的方法,在不同糖質(zhì)量濃度范圍(0.02~0.1、0.1~0.8、0.8~5.0 g/L)內(nèi)寡糖顯色斑點的灰度積分響應值與糖濃度分別有著很好的線性關系(R20.99)。該方法的靈敏度高達0.02 g/L,相對標準偏差為1%~5%,穩(wěn)定性很好。與HPLC等方法相比具有操作簡便、分析快速、不需專門儀器和特定寡糖標準品等優(yōu)點。
[Abstract]:Unidirectional double TLC could improve the analytical effect of thermogel 尾 -1N 3-glucooligosaccharides. The Silica gel60 F254 silica gel plate was used as the stationary phase, V (): V (ethyl acetate): V (water) = 2: 2. 4: 1.4 as the developing agent. When the span is 20 cm, the thermal gel 尾 -1C _ 3-glucooligosaccharides with polymerization degree of 2o _ (13) can be separated by unidirectional twice-unwrapping. In addition, a Image J image analysis method for quantitative thermal gel 尾 -1C 3- glucooligosaccharides was established. There was a good linear relationship between the gray integral response value of oligosaccharide color spots and sugar concentration (R20.99) in different sugar concentration range (0.02 ~ 0.1g / L). The sensitivity of the method is as high as 0.02 g / L, the relative standard deviation is 1 / 5 and the stability is very good. Compared with HPLC and other methods, the method is simple, rapid, and does not require special instruments and specific oligosaccharide standard.
