柯里拉京與DNA相互作用的光譜研究

【摘要】:在p H 7.4的生理條件下,以溴化乙錠(EB)作為熒光探針,利用熒光光譜、紫外-可見吸收光譜、共振散射光譜法結(jié)合鹽效應(yīng)和DNA熔點(T_m)實驗研究了柯里拉京(Cor)與小牛胸腺DNA分子之間的相互作用機制。實驗結(jié)果表明,Cor靜態(tài)猝滅DNA-EB體系的熒光。Cor與DNA作用后,其特征吸收峰強度發(fā)生減色效應(yīng);與DNA作用導致Cor在480.5 nm處的共振散射峰增強,并在330.2 nm處出現(xiàn)新共振散射峰。鹽效應(yīng)對Cor與DNA分子相互作用的影響較小。與Cor作用引起DNA的T_m值升高5.5℃。由此推斷,Cor與DNA相互作用的主要方式為嵌插,兩者間形成了超分子體系。通過計算獲得Cor與DNA間結(jié)合常數(shù)(K_A)為5.82×10~3L/mol(298 K)、2.47×10~4L/mol(310 K),它們之間的作用為熵驅(qū)動的自發(fā)、吸熱過程,疏水作用力是主要的非共價作用方式。
[Abstract]:Under physiological conditions of pH 7.4, ethidium bromide (EB) was used as a fluorescence probe. The mechanism of interaction between Coriragin (Cor) and calf thymus DNA molecule was studied by resonance scattering spectroscopy combined with salt effect and DNA melting point (T _ m) experiment. The experimental results show that the fluorescence of DNA-EB system is quenched by Cor statically, and the characteristic absorption peak intensity of Cor decreases after the interaction of Cor with DNA. The interaction with DNA leads to the enhancement of the resonance scattering peak of Cor at 480.5 nm and the appearance of new resonance scattering peak at 330.2 nm. The salt effect has little effect on the interaction between Cor and DNA molecules. Interaction with Cor resulted in a 5.5 鈩,
