利用UV-C處理研究玫瑰香葡萄中白藜蘆醇及衍生物變化規(guī)律

【摘要】:玫瑰香葡萄是可釀酒可鮮食的葡萄品種,其中含有豐富的白藜蘆醇及其衍生物,它們具有抗氧化、保肝護肝等保健功能。本論文以天津茶淀玫瑰香葡萄為研究對象,旨在研究UV-C照射對玫瑰香葡萄中白藜蘆醇及其衍生物含量的影響。建立了高效液相色譜質(zhì)譜檢測方法,優(yōu)化了提取條件,對比分析了照射劑量、貯藏時間、貯藏溫度對玫瑰香葡萄中白藜蘆醇及其衍生物含量和白藜蘆醇相關(guān)酶酶活力的影響。經(jīng)對比優(yōu)化得到高效液相色譜質(zhì)譜檢測條件為:Thermo Golden(4.6×250 mm,5.0μm)色譜柱;0.1%(v:v)的甲酸水溶液:甲醇體積比20:80等度洗脫;流速0.5mL/min;進樣量10μL,柱溫30℃。ESI-離子掃描模式。3種標準品標準曲線線性關(guān)系良好,相關(guān)系數(shù)均大于0.999,定量限低,精密度高,6次平行測定標準品保留時間相對標準偏差(RSD)小于5%;3個水平加標回收率范圍為85.61%~99.03%,符合實驗要求。在單因素實驗基礎(chǔ)上,以白藜蘆醇、白藜蘆醇苷、紫檀芪提取量總和為響應值,通過響應面法得到最佳工藝條件。結(jié)果表明以乙酸乙酯為提取液,在提取溫度57℃、提取時間32 min、料液比1:15 g/mL條件下,玫瑰香葡萄中芪化物提取量理論值為105.30μg/g,實際提取量可達到104.97μg/g,相對誤差為0.31%。最佳工藝條件可以應用于玫瑰香葡萄中白藜蘆醇、白藜蘆醇苷、紫檀芪的提取。UV-C照射劑量、貯藏時間和貯藏溫度是影響玫瑰香葡萄中白藜蘆醇及其衍生物含量的主要因素。UV-C照射劑量為3.0 kJ/m~2、4.5 kJ/m~24℃貯藏時可以有效抑制玫瑰香葡萄品質(zhì)下降幅度,達到保鮮目的。UV-C照射后立即檢測,劑量為3.0 kJ/m~2時,Res含量達到對照組2.5倍,4.5kJ/m~2時,PD含量達到對照組4倍,PS含量與對照組沒有差別,未能檢出;照射劑量4.5kJ/m~2,4℃避光放置后,Res、PD的含量在48 h內(nèi)到最大值,PS的含量72 h后達到最大值;貯藏溫度為室溫時,48 h后Res和PD含量達到最高90.29μg/g、163.28μg/g;72h后PS含量達到最高46.19μg/g。白藜蘆醇及其衍生物的合成與代謝與相關(guān)酶的酶活力有關(guān)。UV-C照射劑量4.5kJ/m~2時,PAL、β-葡萄糖苷酶酶活力最大;6.0kJ/m~2時,POD、PPO酶活力最大。4.5kJ/m~2照射后4℃避光貯藏24 h后,β-葡萄糖苷酶的酶活力達到最大值,48 h后PAL酶活力達到最大值;96 h后,POD、PPO酶活力達到最大值。對比溫度影響,PAL、β-葡萄糖苷酶、POD、PPO酶活力在室溫時均為最大值15.3 U/gFW、69.5 U/gFW、13.3U/gFW、2.2 U/gFW。經(jīng)對比發(fā)現(xiàn),PAL、β-葡萄糖苷酶酶活力變化趨勢與Res和PD含量的變化趨勢相似,POD、PPO酶活力變化趨勢與PS含量變化趨勢相似,酶活力影響Res的合成與代謝。
