利用UV-C處理研究玫瑰香葡萄中白藜蘆醇及衍生物變化規(guī)律

【摘要】:玫瑰香葡萄是可釀酒可鮮食的葡萄品種,其中含有豐富的白藜蘆醇及其衍生物,它們具有抗氧化、保肝護(hù)肝等保健功能。本論文以天津茶淀玫瑰香葡萄為研究對(duì)象,旨在研究UV-C照射對(duì)玫瑰香葡萄中白藜蘆醇及其衍生物含量的影響。建立了高效液相色譜質(zhì)譜檢測(cè)方法,優(yōu)化了提取條件,對(duì)比分析了照射劑量、貯藏時(shí)間、貯藏溫度對(duì)玫瑰香葡萄中白藜蘆醇及其衍生物含量和白藜蘆醇相關(guān)酶酶活力的影響。經(jīng)對(duì)比優(yōu)化得到高效液相色譜質(zhì)譜檢測(cè)條件為:Thermo Golden(4.6×250 mm,5.0μm)色譜柱;0.1%(v:v)的甲酸水溶液:甲醇體積比20:80等度洗脫;流速0.5mL/min;進(jìn)樣量10μL,柱溫30℃。ESI-離子掃描模式。3種標(biāo)準(zhǔn)品標(biāo)準(zhǔn)曲線線性關(guān)系良好,相關(guān)系數(shù)均大于0.999,定量限低,精密度高,6次平行測(cè)定標(biāo)準(zhǔn)品保留時(shí)間相對(duì)標(biāo)準(zhǔn)偏差(RSD)小于5%;3個(gè)水平加標(biāo)回收率范圍為85.61%~99.03%,符合實(shí)驗(yàn)要求。在單因素實(shí)驗(yàn)基礎(chǔ)上,以白藜蘆醇、白藜蘆醇苷、紫檀芪提取量總和為響應(yīng)值,通過(guò)響應(yīng)面法得到最佳工藝條件。結(jié)果表明以乙酸乙酯為提取液,在提取溫度57℃、提取時(shí)間32 min、料液比1:15 g/mL條件下,玫瑰香葡萄中芪化物提取量理論值為105.30μg/g,實(shí)際提取量可達(dá)到104.97μg/g,相對(duì)誤差為0.31%。最佳工藝條件可以應(yīng)用于玫瑰香葡萄中白藜蘆醇、白藜蘆醇苷、紫檀芪的提取。UV-C照射劑量、貯藏時(shí)間和貯藏溫度是影響玫瑰香葡萄中白藜蘆醇及其衍生物含量的主要因素。UV-C照射劑量為3.0 kJ/m~2、4.5 kJ/m~24℃貯藏時(shí)可以有效抑制玫瑰香葡萄品質(zhì)下降幅度,達(dá)到保鮮目的。UV-C照射后立即檢測(cè),劑量為3.0 kJ/m~2時(shí),Res含量達(dá)到對(duì)照組2.5倍,4.5kJ/m~2時(shí),PD含量達(dá)到對(duì)照組4倍,PS含量與對(duì)照組沒(méi)有差別,未能檢出;照射劑量4.5kJ/m~2,4℃避光放置后,Res、PD的含量在48 h內(nèi)到最大值,PS的含量72 h后達(dá)到最大值;貯藏溫度為室溫時(shí),48 h后Res和PD含量達(dá)到最高90.29μg/g、163.28μg/g;72h后PS含量達(dá)到最高46.19μg/g。白藜蘆醇及其衍生物的合成與代謝與相關(guān)酶的酶活力有關(guān)。UV-C照射劑量4.5kJ/m~2時(shí),PAL、β-葡萄糖苷酶酶活力最大;6.0kJ/m~2時(shí),POD、PPO酶活力最大。4.5kJ/m~2照射后4℃避光貯藏24 h后,β-葡萄糖苷酶的酶活力達(dá)到最大值,48 h后PAL酶活力達(dá)到最大值;96 h后,POD、PPO酶活力達(dá)到最大值。對(duì)比溫度影響,PAL、β-葡萄糖苷酶、POD、PPO酶活力在室溫時(shí)均為最大值15.3 U/gFW、69.5 U/gFW、13.3U/gFW、2.2 U/gFW。經(jīng)對(duì)比發(fā)現(xiàn),PAL、β-葡萄糖苷酶酶活力變化趨勢(shì)與Res和PD含量的變化趨勢(shì)相似,POD、PPO酶活力變化趨勢(shì)與PS含量變化趨勢(shì)相似,酶活力影響Res的合成與代謝。

It has the function of antioxidation, protecting liver and so on. In this paper, the effect of UV-C irradiation on resveratrol and its derivatives was studied.
