辣木葉的HPLC指紋圖譜

【摘要】:目的:通過建立辣木葉的HPLC指紋圖譜,評價不同產(chǎn)地辣木葉質(zhì)量的差異,為辣木質(zhì)量控制和評價提供科學(xué)依據(jù),推動辣木的開發(fā)與利用。方法:采用HPLC法,Diamonsil Plus C_(18)色譜柱(4.6 mm×250 mm,5μm),流動相乙腈(A)-0.05%磷酸溶液(B)(0~70 min,5%~30%A;70~75 min,30%~100%A),流速1.0 mL·min~(-1),檢測波長230 nm,柱溫30℃。測定5個不同產(chǎn)地12批辣木葉的指紋圖譜,并采用國家藥典委員會頒布的"中藥色譜指紋圖譜相似度評價系統(tǒng)"(2004 A)和SPSS 21.0統(tǒng)計軟件中的聚類分析對指紋圖譜進(jìn)行相似度比較。結(jié)果:建立辣木葉指紋圖譜共有模式,標(biāo)定14個指紋圖譜共有峰,其中3個成分峰分別為沒食子酸、蘆丁、槲皮苷,為其有效成分。12批辣木葉中9批的相似度0.90,說明不同產(chǎn)地及同一產(chǎn)地不同地區(qū)辣木葉指紋圖譜既有共性又存在一定差異。結(jié)論:建立的方法精密度、重復(fù)性和穩(wěn)定性較好,可為辣木葉的鑒別和質(zhì)量評價提供參考。
