川牛膝酒炙和鹽炙前后HPLC化學(xué)指紋圖譜及其主要藥效成分量變化研究

【摘要】:目的揭示川牛膝藥材經(jīng)酒炙和鹽炙前后的化學(xué)成分變化規(guī)律,為修訂和完善川牛膝質(zhì)量標(biāo)準(zhǔn)提供參考。方法采用Agilent Zorbax Eclipse XDB-C18(250 mm×4.6 mm,5μm)色譜柱,0.1%磷酸水溶液-乙腈為流動(dòng)相梯度洗脫,檢測波長266 nm,體積流量0.5 m L/min,建立HPLC指紋圖譜,記錄各色譜峰的保留時(shí)間和峰面積數(shù)據(jù)。采用對(duì)照品指認(rèn)主要色譜峰物質(zhì),采用向量夾角法計(jì)算指紋圖譜相似系數(shù),采用主成分分析和聚類分析方法進(jìn)行模式識(shí)別。結(jié)果從24批次樣品中一共提取出25個(gè)主要色譜峰,指認(rèn)了其中的葛根素、杯莧甾酮、大豆苷元3個(gè)色譜峰。主成分分析、聚類分析以及相似度結(jié)果顯示,酒炙和鹽炙對(duì)川牛膝化學(xué)成分的整體組成與其量水平具有顯著影響。葛根素、杯莧甾酮、大豆苷元3個(gè)已知的藥效成分在經(jīng)酒炙后其量均未得到顯著提升,經(jīng)鹽炙后僅有葛根素的量得到一定程度地提升。結(jié)論現(xiàn)行藥效指標(biāo)成分對(duì)于川牛膝酒炙品和鹽炙品的質(zhì)控作用較弱,有關(guān)川牛膝炮制品的質(zhì)控指標(biāo)成分還需進(jìn)一步篩選。實(shí)驗(yàn)所建立的HPLC化學(xué)指紋圖譜及其分析方法可以用于川牛膝生品、酒炙品、鹽炙品之間的鑒定及其質(zhì)量控制。
[Abstract]:Objective to reveal the changes of chemical constituents of Radix Achyranthes bidentata before and after liquor and salt processing, and to provide a reference for revising and improving the quality standard of Radix Achyranthes bidentata. Methods Agilent Zorbax Eclipse XDB-C18 (250 mm 脳 4.6 mm,5 渭 m) column, 0.1% phosphoric acid aqueous solution and acetonitrile as mobile phase gradient elution, detection wavelength 266 nm, volume flow rate 0.5 mL / min, established HPLC fingerprint, recorded the retention time and peak area data of each chromatographic peak. The main chromatographic peak substances were identified by reference substance, the similarity coefficient of fingerprint was calculated by vector angle method, and the pattern recognition was carried out by principal component analysis and cluster analysis. Results A total of 25 main chromatographic peaks were extracted from 24 batches of samples, and three peaks of puerarin, calix amaranone and daidzein were identified. The results of principal component analysis, cluster analysis and similarity analysis showed that liquor and salt processing had significant effects on the composition and quantity of the chemical constituents of Achyranthes bidentata. Puerarin, calix amaranone and daidzein had no significant increase in the amount of puerarin, but only puerarin increased to a certain extent. Conclusion the quality control of Radix Achyranthes bidentata Radix angelicae officinalis and salt-broiled products is weak, and further screening is needed for the quality control index components of processed products of Radix Achyranthes bidentata. The HPLC fingerprint and its analytical method can be used for the identification and quality control of Radix Achyranthes bidentata, Radix angelicae Salina and Radix Achyranthes bidentata.
