延胡索HPLC指紋圖譜研究及9種生物堿含量測定

【摘要】:目的:分別建立延胡索藥材及醋延胡索飲片的HPLC指紋圖譜,同時對其中9種生物堿成分進行含量測定。方法:以通過三乙胺調節(jié)pH值至6.1的乙腈-0.1%磷酸溶液作為流動相,選擇ECOSIL 120-5-C18AQ色譜柱(250 mm×4.6 mm,5.0μm)進行梯度洗脫。結果:延胡索藥材和醋延胡索飲片各10批分別所得的指紋圖譜,相似度值均0.94,確定共有峰均有15個。原阿片堿、鹽酸巴馬汀、鹽酸小檗堿、脫氫紫堇堿、海罌粟堿、延胡索乙素、四氫小檗堿、延胡索甲素、脫氫海罌粟堿9種生物堿在相應范圍內與其峰面積均呈良好線性關系(r≥0.9999);其平均加樣回收率(n=6)分別為99.0%、101.5%、99.0%、102.2%、98.7%、100.3%、101.8%、99.4%、99.8%,RSD≤2.5%。結論:該方法具有較高的靈敏度,良好的重復性,可為延胡索藥材和醋延胡索飲片的質量評價和全面控制提供參考依據。
