高良姜等五味山姜屬中藥乙酸乙酯部位的HPLC指紋圖譜

【摘要】:以95%乙醇作為提取的溶媒,將加熱回流提取得到的提取液水浴揮干得到浸膏,并用甲醇配制成1 mg·m L~(-1)供試液。采用phenomenex-C18(250 mm×4.6 mm,5μm)色譜柱,流動(dòng)相為乙腈-0.1%磷酸水溶液,梯度洗脫,流速為1.0 m L·min~(-1),柱溫為30℃,檢測波長為260 nm HPLC色譜法測定,采用國家藥典委員會(huì)相似度評價(jià)軟件進(jìn)行分析。結(jié)果表明:建立了精密度、穩(wěn)定性和重現(xiàn)性均較好的5味山姜屬中藥乙酸乙酯部位HPLC指紋圖譜,確定了9個(gè)共有峰,6個(gè)強(qiáng)峰;其中高良姜、大高良姜含有5個(gè)共有峰,高良姜和草豆蔻含有3個(gè)共有峰,大高良姜和紅豆蔻含有2個(gè)共有峰,高良姜和益智含有3個(gè)共有峰。對5味山姜屬中藥乙酸乙酯部位化學(xué)成分進(jìn)行相似度分析,得出其相似度分別為0.955、0.805、0.371、0.794、0.345。所建立的5味山姜屬中藥乙酸乙酯部位的指紋圖譜方法穩(wěn)定、簡便、可靠,其特征峰所代表的化學(xué)成分的相似性不盡相同,高良姜、大高良姜、草豆蔻的相似性較大,紅豆蔻、益智的相似性較小,說明同基原中藥之間的化學(xué)成分有一定的相關(guān)性。該研究結(jié)果為探討山姜屬中藥親緣關(guān)系與化學(xué)成分相關(guān)性提供了思路與基礎(chǔ)。
