高效液相色譜-示差折光率法測定林業(yè)生物質(zhì)原料中的可溶性糖

【摘要】:建立了高效液相色譜測定林業(yè)生物質(zhì)原料中可溶性糖的含量和種類的方法。前處理方法為樣品稱樣量4~5g,蒸餾水熱浸提1.0 h。色譜條件為:色譜柱(XBridge Amide NUCLEOSIL100-5 NH24.6×250 mm,3.5μm);流動相為V(乙腈):V(水)=83:17,加入0.1%(V/V)的三乙胺;柱溫為30℃;流速1.0 m L/min;進樣量20μL。在此條件下,葡萄糖、木糖、阿拉伯糖、果糖、甘露糖和蔗糖6種糖的質(zhì)量濃度分別與峰面積呈良好的線性關系(R20.9978),檢測限在3.26~4.30 mg/L之間;加標回收率在96.2%~103.4%之間。對毛竹竹材,油茶餅粕,橡膠樹樹干,虎榛子樹干,胡楊樹干,白樺根、枝、樹干,馬尾松針葉,銀杏葉,毛白楊樹干等實際樣品進行測定時發(fā)現(xiàn),林木生物質(zhì)的種類、取樣部位不同,所含有的可溶性糖種類及含量差異較大。該方法可用于測定林業(yè)生物質(zhì)原料中可溶性糖含量。
