具有可控質(zhì)子化能力的陽(yáng)離子聚合物的合成及表征

【摘要】:合成了聚乙二醇-聚賴氨酸嵌段共聚物(PEG-PLL),并利用Michael加成反應(yīng)對(duì)其側(cè)鏈進(jìn)行叔胺和季銨化修飾,得到一系列具有支化胺結(jié)構(gòu)和可控質(zhì)子化能力的聚賴氨酸衍生物。通過核磁共振氫譜、傅里葉變換紅外光譜和凝膠滲透色譜對(duì)聚合物的結(jié)構(gòu)進(jìn)行表征。利用pH滴定研究聚合物的質(zhì)子化行為,發(fā)現(xiàn)季銨和叔胺修飾能夠顯著改變陽(yáng)離子聚合物的質(zhì)子緩沖能力,將pKa值從10.6分別降低至7.5和6.8。圓二色譜測(cè)試結(jié)果表明側(cè)基修飾不僅使聚賴氨酸嵌段從無規(guī)卷曲構(gòu)象變?yōu)楹笑谅菪挺抡郫B的混合構(gòu)象,而且能夠調(diào)節(jié)其高級(jí)結(jié)構(gòu)對(duì)腫瘤微環(huán)境的響應(yīng)能力。
[Abstract]:Poly (ethylene glycol) -poly (lysine) block copolymer (PEG-PLL) was synthesized and modified with tertiary amine and quaternary ammonium by Michael addition reaction. A series of poly (lysine) derivatives with branched amine structure and controllable protonation ability were obtained. The structure of the polymer was characterized by nuclear magnetic resonance (NMR), Fourier transform infrared spectroscopy (FTIR) and gel permeation chromatography (GPC). PH titration was used to study the protonation behavior of cationic polymers. It was found that the modification of quaternary ammonium and tertiary amine could significantly change the proton buffer capacity of cationic polymers, reducing the pKa value from 10.6 to 7.5 and 6.8 respectively. The results of circular dichroism showed that the modification not only changed the poly (lysine) block from random coil conformation to a mixed conformation with 偽 helix and 尾 folding, but also regulated the response of its advanced structure to tumor microenvironment.
