中藥成分香草酸和咖啡酸的熒光分析方法研究

【摘要】:本文研究了中藥活性成分香草酸、咖啡酸、咖啡酸乙酯的熒光性質(zhì),探討了中藥材胡黃連、中成藥肥兒丸、黃連羊肝丸中的香草酸以及中藥材蒲公英中的咖啡酸的定量分析方法。實(shí)驗(yàn)中,用三維熒光技術(shù)獲取標(biāo)準(zhǔn)溶液和樣品溶液全面的熒光信息,通過比較三維熒光圖譜的一致性對(duì)樣品中可能存在的干擾做出初步判斷,然后采用熒光分光光度法和基于交替三線性分解的二階校正技術(shù)處理光譜數(shù)據(jù),以選擇合適的分析方法。論文工作主要包括以下三部分:1、研究了香草酸的熒光性質(zhì)。香草酸內(nèi)源熒光很弱,但在香草酸水溶液中加入AlCl3可使熒光明顯增強(qiáng),此熒光在弱酸性條件下穩(wěn)定,最大激發(fā)波長(zhǎng)λex為260 nm,最大發(fā)射波長(zhǎng)λem為344 nm。探究了實(shí)驗(yàn)條件對(duì)香草酸敏化熒光的影響。以L-色氨酸為參比,測(cè)得香草酸-Al(Ⅲ)絡(luò)合物的熒光量子產(chǎn)率為0.21,屬于強(qiáng)熒光物質(zhì)。2、基于香草酸的熒光性質(zhì),用熒光分光光度法測(cè)定了中藥材胡黃連、中成藥肥兒丸、黃連羊肝丸香草酸的含量,測(cè)定結(jié)果為:胡黃連含香草酸0.434%、肥兒丸含香草酸0.106%,黃連羊肝丸含香草酸0.0344%。用三維熒光二階校正法對(duì)上述樣品進(jìn)行測(cè)定,結(jié)果分別為:0.373%、0.101%和0.0303%。通過對(duì)比可見,兩種方法對(duì)肥兒丸的測(cè)定結(jié)果基本一致,但對(duì)胡黃連和黃連羊肝丸的測(cè)定不同,表明三維熒光二階校正法對(duì)共存組分的干擾有一定的分辨能力。3、研究了咖啡酸和咖啡酸乙酯的熒光性質(zhì),探究了光照對(duì)咖啡酸熒光強(qiáng)度的影響,發(fā)現(xiàn)光照能使咖啡酸的熒光增強(qiáng)。用熒光分光光度法對(duì)蒲公英中的咖啡酸進(jìn)行定量分析,測(cè)定結(jié)果為0.0232%,用三維熒光二階校正法測(cè)定結(jié)果為0.0240%,兩者基本一致。本節(jié)還探討了用三維熒光二階校正法對(duì)咖啡酸和咖啡酸乙酯混合溶液中兩種組分的含量分別進(jìn)行測(cè)定的可行性,得到了有意義的結(jié)果。
[Abstract]:The fluorescence properties of vanillic acid and ethyl caffeic acid were studied in this paper. Quantitative analysis of vanillic acid in Huanglian Yanggan pills and caffeic acid in dandelion. In the experiment, the comprehensive fluorescence information of standard solution and sample solution was obtained by using three-dimensional fluorescence technique, and the possible interference in the sample was preliminarily judged by comparing the consistency of the three-dimensional fluorescence spectrum. Then the fluorescence spectrophotometry and the second-order correction technique based on alternating trilinear decomposition were used to process the spectral data in order to select an appropriate analytical method. In this paper, the fluorescence properties of vanillic acid were studied in the following three parts: 1. The endogenous fluorescence of vanillic acid is very weak, but the fluorescence of vanillic acid aqueous solution can be enhanced by adding AlCl3. The fluorescence is stable under weak acid condition. The maximum excitation wavelength 位 ex is 260 nm and the maximum emission wavelength 位 em is 344 nm.
