兩種氧化還原體系引發(fā)的整體柱的制備及性能研究

【摘要】:聚合物整體柱因具有制備方法簡(jiǎn)單、傳質(zhì)快、滲透性好等優(yōu)點(diǎn),而被廣泛應(yīng)用于生命科學(xué)、環(huán)境科學(xué)和生物科學(xué)等領(lǐng)域的色譜分離分析中。本文通過采用氧化還原體系引發(fā)制備了兩種聚合物整體柱,旨在改善聚合物整體柱分離小分子化合物的柱效和提高其對(duì)于生物大分子的選擇性。首先以二甲基丙烯酸-1,6-己二醇酯作為單體,乙二醇二甲基丙烯酸酯為交聯(lián)劑,環(huán)己醇和1,4-丁二醇為二元致孔劑,過氧化苯甲酰和N,N-二甲基苯胺組成的氧化還原體系為引發(fā)劑,在30℃水浴下成功地制備了聚合物整體柱。分別通過掃描電子顯微鏡、氮吸附-脫附法、壓汞法對(duì)該整體柱進(jìn)行了表征,結(jié)果表明該整體柱具有孔徑分布較均勻的多孔骨架結(jié)構(gòu)。高效液相色譜實(shí)驗(yàn)表明,將該整體柱作為固定相,成功地分離了七種芳香族化合物的混合物,其中柱效最高可達(dá)21,000理論塔板/米。同時(shí)對(duì)掌葉大黃中的五種活性成分進(jìn)行了分離和定量分析,方法的回收率在98.48%至101.57%之間,五種活性成分的檢測(cè)限和定量限分別小于33.7 ng和90.8 ng。其次,采用相同的引發(fā)體系,以氯化血紅素和甲基丙烯酸月桂酯為二元單體;乙二醇二甲基丙烯酸酯為交聯(lián)劑;正丁醇、乙二醇、N,N-二甲基甲酰胺為三元致孔劑;在室溫下制備了聚合物整體柱。通過掃描電子顯微鏡、氮吸附-脫附法、壓汞法探究了該整體柱的內(nèi)部孔結(jié)構(gòu),表明該整體柱具有均勻的多孔結(jié)構(gòu)。以該整體柱作為高效液相色譜固定相,對(duì)五種芳香族化合物的混合物進(jìn)行了分離,最高柱效可達(dá)19,880理論塔板/米。此外,該整體柱對(duì)于復(fù)雜的生物樣品,如人血漿、雞卵清、蝸牛酶等中的蛋白質(zhì)具有良好的選擇性和分離能力。值得一提的是,該整體柱能夠在線去除人血漿中的三種高豐度蛋白,并同時(shí)對(duì)人血漿中的其他蛋白質(zhì)進(jìn)行分級(jí)分離,展現(xiàn)了氯化血紅素對(duì)于改善整體柱對(duì)蛋白質(zhì)的選擇性的重要作用。以上實(shí)驗(yàn)結(jié)果表明,本文通過氧化還原引發(fā)體系制備的整體柱能夠在一定程度上提高分離小分子化合物的柱效,基于氯化血紅素的整體柱則較大程度的改善了聚合物整體柱對(duì)于復(fù)雜生物基質(zhì)中蛋白質(zhì)的選擇性。
