UPC~2-MS法快速檢測蜂蜜中的果糖、葡萄糖、蔗糖和麥芽糖

【摘要】:建立了蜂蜜中果糖、葡萄糖、蔗糖和麥芽糖的超高效合相色譜-質(zhì)譜(UPC2-MS)快速檢測的方法。樣品先用少量的水溶解,再用正己烷/異丙醇(1:1,V/V)溶解,經(jīng)ACQUITY UPC2BEH(100 mm×3.0 mm,1.7μm)色譜柱分離,以超臨界CO2-甲醇(含0.1%NH3·H2O,V/V)為流動相進(jìn)行梯度洗脫,流速為1.8 m L/min,通過質(zhì)譜檢測器在負(fù)離子電噴霧模式下對目標(biāo)化合物進(jìn)行分析,外標(biāo)法定量。檢測結(jié)果表明:果糖、葡萄糖、蔗糖和麥芽糖在2~150 mg/L范圍線性關(guān)系良好,R20.9990,定量限(LOQ,S/N≥10)為0.22~0.42 mg/L,高、中、低3個濃度水平的加標(biāo)平均回收率在90.0%~107.5%范圍,相對標(biāo)準(zhǔn)偏差(RSD,n=6)均小于4.0%。該方法簡單快速,分離效果好,為蜂蜜中果糖、葡萄糖、蔗糖和麥芽糖的分離測定提供了新的色譜技術(shù)平臺。
[Abstract]:A rapid method for the determination of fructose, glucose, sucrose and maltose in honey by superhigh performance phase chromatography-mass spectrometry (UPC2-MS) was established. The sample was dissolved in a small amount of water, then dissolved with n-hexane / isopropanol (1: 1 V / V). The sample was separated by ACQUITY UPC2BEH (100mm 脳 3.0mm ~ (1.7) 渭 m) column. The supercritical CO _ 2-methanol (containing 0.1%NH3 H _ 2O _ 2O _ V) was used as the mobile phase for gradient elution. At a flow rate of 1.8 mL / min, the target compounds were analyzed by mass spectrometer in anion electrospray mode and quantified by external standard method.
