大孔吸附樹脂分光光度法檢測瑪咖總皂苷含量

【摘要】:探討瑪咖中總皂苷含量測定的方法。以《保健食品檢驗與評價技術(shù)規(guī)范(2003版)》中"保健食品中總皂苷的測定"為基礎(chǔ),考察用D101大孔吸附樹脂代替Amberlite-XAD-2大孔樹脂純化總皂苷的可行性,并研究樣品在樹脂中的吸附時間、樣品總皂苷的濃度兩方面對檢測結(jié)果的影響。結(jié)果表明,標準曲線在0~0.176 mg/m L范圍內(nèi)線性關(guān)系良好,線性回歸方程y=3.308 4x-0.006 8(R2=0.999 0),方法檢出限為2μg/m L。兩種樹脂的總皂苷檢測結(jié)果無顯著差異。樣品吸附時間為60 min,樣品中總皂苷含量在0.08~0.32 mg/m L范圍內(nèi)時,總皂苷的檢測誤差小。改進后的方法平均加標回收率99.76%,RSD為2.23%(n=6),精密度試驗RSD為2.97%(n=6)。改進后的總皂苷檢測方法靈敏度高,準確性好,重現(xiàn)性好,可用于測定瑪咖中總皂苷的含量,瑪咖中總皂苷含量為3.81±0.11 mg/g(n=6)。
[Abstract]:A method for the determination of total saponins in Maca was studied. Based on "determination of Total saponins in Health Foods" (2003 Edition), the feasibility of purifying total saponins by D101 macroporous resin instead of Amberlite-XAD-2 macroporous resin was investigated. The effects of adsorption time and concentration of total saponins on the results were studied. The results show that the standard curve has a good linear relationship in the range of 0 ~ 0.176 mg/m / L, and the linear regression equation is 3.308 4x-0.006 _ 8 (R _ 2N _ (0.999). The detection limit of the method is 2 渭 g / m ~ (L). There was no significant difference in total saponins between the two resins. When the adsorption time was 60 minutes and the content of total saponins was in the range of 0.08 ~ 0.32 mg/m / L, the detection error of total saponins was small. The average recovery of the improved method was 99.76% (RSD = 2.23%) and the precision test (RSD) was 2.97% (n = 6). The improved method has high sensitivity, accuracy and reproducibility. It can be used to determine the content of total saponins in Maca. The content of total saponins in Maca is 3.81 鹵0.11 mg/g (NQ 6).
