高效液相色譜法測定牛羊雜碎等肉類中嘌呤及尿酸

【摘要】:建立同時測定肉類食品中尿酸、鳥嘌呤、次黃嘌呤、黃嘌呤和腺嘌呤的高效液相色譜分析方法,并測定肉類樣品中上述4種嘌呤及尿酸的含量。色譜條件:資生堂CAPCELL PAK-C_(18)(4.6 mm×250 mm,5μm)色譜柱,柱溫30℃;流動相為7×10~(-3)mol/L KH_2PO_4-H_3PO_4(pH=3.83),流速1.0 mL/min;檢測器為紫外檢測器,檢測波長254 nm;進(jìn)樣量10μL。結(jié)果:在上述條件下4種嘌呤和尿酸分離和測定效果良好,4種嘌呤和尿酸的質(zhì)量濃度和峰面積在0.05~50μg/mL線性范圍內(nèi)線性關(guān)系良好,相關(guān)系數(shù)均在0.999 6以上,檢出限在0.010~0.024μg/mL之間,精密度檢測RSD為0.25%~0.81%,各組分回收率為92.0%~105.0%,方法精密度RSD為6.27%~11.09%。結(jié)論:該方法簡便,快捷,可靠,各組分分離度好,可應(yīng)用于肉類食品中嘌呤和尿酸的同時分析測定。
