應用高效液相色譜-二極管陣列檢測技術(shù)測定阿卡波糖片含量的研究

【摘要】:目的應用高效液相色譜-二極管陣列檢測(HPLC-DAD)技術(shù),建立阿卡波糖片中阿卡波糖的含量測定方法。方法采用Alltima C_(18)色譜柱(4.6 mm×250 mm,5μm),以乙腈~(-1)0 mmol·L~(-1)磷酸二氫銨溶液(含0.04%辛烷磺酸鈉,磷酸調(diào)至pH 3.3)(15∶85)為流動相,流速0.8 m L·min~(~(-1)),檢測波長200 nm,柱溫30℃。結(jié)果在本方法中,阿卡波糖的線性關(guān)系良好,線性相關(guān)系數(shù)(r)為1.000 0;專屬性強,阿卡波糖和藥品中其他成分以及強制降解產(chǎn)物之間均有良好的分離度;精密度和穩(wěn)定性良好,RSD分別為0.088%和0.22%;回收率良好,3個加樣濃度的平均回收率(n=9)為98.49%,RSD為0.24%。對4批阿卡波糖片進行測定,含量分別為每片51.69、51.75、51.72、51.62 mg。結(jié)論該方法準確、簡單、快速,可作為阿卡波糖片中阿卡波糖的含量測定方法。
