熒光增強型金屬納米簇的生物檢測研究

【摘要】:近些年來,在催化,傳感等領(lǐng)域,金屬納米材料被證實有著廣泛的應(yīng)用?茖W(xué)家們發(fā)現(xiàn),腺嘌呤及其衍生物可以被用來合成納米顆粒,然而利用它們合成的配合物、聚合物和納米顆粒之中,僅僅有非常少的一部分產(chǎn)物可以產(chǎn)生熒光。在本論文中,我們選取了金納米材料與腺苷來合成AuAXP(單磷酸腺苷、二磷酸腺苷、三磷酸腺苷)作為實驗研究基礎(chǔ)。首先,將腺苷和金納米顆粒結(jié)合,觀察所得到的產(chǎn)物各自的熒光強度。然后進行對照實驗,比較在不同添加物影響下產(chǎn)物的熒光增強效應(yīng),比如加入鈣、鎂、鋅等離子會否發(fā)生增強作用,并且考察觀察在熒光增強作用下,原本的產(chǎn)物在細胞成像方面的表現(xiàn)有什么變化。本課題主要對熒光增強型金納米簇的制備和表征,以及其在細胞和生物分子中的檢測和成像進行了研究,主要利用熒光光譜儀、共聚焦熒光顯微鏡、紫外光譜儀以及酶標儀等手段重點研究了不同熒光金納米簇在細胞和生物分子檢測和生物成像中的作用,并對其潛在的應(yīng)用前景進行展望。本實驗選取了氯金酸溶液作為反應(yīng)前驅(qū)體,檸檬酸作為還原劑和保護劑,探究了在不同的條件下例如不同濃度,不同溫度的反應(yīng)前驅(qū)體,對合成產(chǎn)物納米金簇的影響。并從結(jié)果之中對比挑選了最優(yōu)的方案,成功制備了熒光金納米簇。本實驗的這種合成方法操作簡便,實驗所需成本相對較低,所合成的金納米簇具有低熒光性、熒光壽命長、顆粒粒徑小且粒徑分布十分均一、生物相容性好無毒性等等優(yōu)點,且在腺苷酸衍生物上,具有許多的能與金屬離子結(jié)合的位點,極為有效的增強了熒光強度。在本論文中,我們通過考察了不同金屬離子對合成的熒光金屬復(fù)合物的熒光增強效應(yīng)和生物成像能力,不僅僅證明了熒光增強型金納米簇在生物檢測,細胞成像等等方面都有著有效的應(yīng)用,還為如何提高檢測的精確度,成像清晰度等至關(guān)重要的要素提供了一些不同的研究思路。
[Abstract]:In recent years, metal nanomaterials have been widely used in catalysis, sensing and other fields. Scientists have found that adenine and its derivatives can be used to synthesize nanoparticles, but very few of the complexes, polymers and nanoparticles they synthesize produce fluorescence. In this thesis, we selected gold nanomaterials and adenosine to synthesize AuAXP (adenosine monophosphate, adenosine diphosphate, adenosine triphosphate) as the experimental basis. Firstly, the fluorescence intensity of the product was observed by combining adenosine with gold nanoparticles. Then the contrast experiment was conducted to compare the fluorescence enhancement effect of the product under the influence of different additives, such as calcium, magnesium and zinc plasma, and to observe the effect of fluorescence enhancement. What changes have been made in the performance of the original product in cellular imaging. In this paper, the preparation and characterization of fluorescent enhanced gold nanoclusters and their detection and imaging in cells and biomolecules were studied, mainly using fluorescence spectrometer and confocal fluorescence microscope. The role of different fluorescent gold nanoclusters in cell and biomolecular detection and biomolocation was studied by means of UV spectrometer and enzyme labeling. The potential applications of these clusters were also prospected. In this experiment, chlorgold acid solution was selected as the reaction precursor, citric acid as reducing agent and protective agent. The effects of different concentration and temperature of reaction precursor on the synthesis of nano-gold clusters were investigated. The optimal scheme was selected from the results, and the fluorescent gold nanoclusters were successfully prepared. The synthesized gold nanoclusters have the advantages of low fluorescence, long fluorescence life, small particle size and uniform particle size distribution, good biocompatibility, etc. In addition, adenylic acid derivatives have many sites that can bind to metal ions, which can effectively enhance the fluorescence intensity. In this thesis, we investigated the fluorescence enhancement effect and the biological imaging ability of the fluorescent metal complexes synthesized by different metal ions, which not only proved that the fluorescence enhanced gold nanoclusters were in the biological detection. Cell imaging and other aspects of effective applications, but also for how to improve the accuracy of detection, imaging clarity and other crucial elements to provide some different research ideas.
