殼聚糖氧化降解制備聚合度6以下的殼寡糖

【摘要】:殼寡糖是殼聚糖經(jīng)降解后的低聚物,其相對分子質(zhì)量較小,水溶性好,易于吸收,生物活性高。本實(shí)驗(yàn)采用過氧化氫氧化降解脫乙酰度高于95%的殼聚糖,利用離子色譜-脈沖安培法對低聚合度的殼寡糖進(jìn)行定性和定量分析,以制備聚合度在6以下的殼寡糖。以降解產(chǎn)物的離子色譜中聚合度6以下的峰面積和與產(chǎn)物得率之積作為響應(yīng)指標(biāo),并結(jié)合凝膠滲透色譜來考察過氧化氫濃度、反應(yīng)時(shí)間、反應(yīng)溫度對殼聚糖降解的影響及降解特性,并利用響應(yīng)面分析法對氧化條件進(jìn)行優(yōu)化。研究結(jié)果表明,采用離子色譜測定聚合度6以下的殼寡糖在方法學(xué)上是可行的,具有良好的精密度、穩(wěn)定性和重現(xiàn)性。得到的最佳降解工藝條件為:過氧化氫濃度4.50%、反應(yīng)時(shí)間6 h、反應(yīng)溫度56℃。由最優(yōu)條件下的降解產(chǎn)物離子色譜圖可知,聚合度越低的殼寡糖越容易得到;可利用離子色譜法對殼聚糖的降解過程進(jìn)行調(diào)控。
[Abstract]:Chitosan oligosaccharides are low molecular weight, water soluble, easy to absorb and high bioactivity. The chitosan whose degree of deacetylation is more than 95% was oxidized by hydrogen peroxide. The chitosan with low degree of polymerization was prepared by ion chromatography-pulse amperometric method. The peak area of polymerization degree below 6 and the product of product yield were used as the response index, and the concentration of hydrogen peroxide and reaction time were investigated by gel permeation chromatography. The effect of reaction temperature on the degradation of chitosan and its degradation characteristics were studied. The oxidation conditions were optimized by response surface analysis (RSM). The results show that the determination of chitosan oligosaccharides with polymerization degree below 6 by ion chromatography is feasible in methodology and has good precision, stability and reproducibility.
