基于UPLC-Q-TOF-MS技術(shù)的廣地龍血清藥物化學(xué)初步研究

【摘要】:目的:對廣地龍的血清藥物化學(xué)進(jìn)行初步研究。方法:應(yīng)用超高液相色譜-串聯(lián)四級桿飛行時(shí)間質(zhì)譜技術(shù)(UPLC-Q-TOF-MS)建立廣地龍及其含藥血清樣品的分析方法;比較體外樣品、空白血清及含藥血清分析結(jié)果,結(jié)合Peakview 2.0和Metabolite Pilot 1.5數(shù)據(jù)分析軟件鑒定和表征廣地龍血中移行成分。結(jié)果:從大鼠含藥血清中發(fā)現(xiàn)了13個(gè)移行成分,包括10個(gè)原型成分和3個(gè)代謝產(chǎn)物。結(jié)論:初步確定了廣地龍的血中原型成分及代謝產(chǎn)物,有助于闡明其藥效物質(zhì)基礎(chǔ)和作用機(jī)理。
[Abstract]:Objective: to study the serum pharmacochemistry of Guandelong. Methods: UPLC-Q-TOF-MS was used to establish a method for the analysis of Guandelong and its drug-containing serum samples, and to compare the analysis results of in vitro samples, blank serum and drug-containing serum. Peakview 2.0 and Metabolite pilot 1.5 data analysis software were used to identify and characterize the transitory components in the blood of Dioscorea grandii. Results: thirteen transitory components including 10 prototypes and 3 metabolites were found in rat serum containing drugs. Conclusion: the prototypes and metabolites in the blood of Dioscorea grandifolia were preliminarily determined, which was helpful to elucidate the substance basis and mechanism of its pharmacodynamics.
