密蒙花中4個化合物含量的多波長高效液相色譜法測定

【摘要】:目的建立HPLC多波長法測定密蒙花中4個化合物(毛蕊花糖苷、蒙花苷、木犀草素、芹菜素)的含量,為密蒙花藥材質(zhì)量標(biāo)準(zhǔn)的建立提供參考。同時對不同產(chǎn)地密蒙花進(jìn)行比較研究。方法采用高效液相色譜法,使用Waters Symmetry Columns(4.6mm×250mm,5 um)色譜柱,以甲醇(A)-0.1%磷酸水(B)為流動相,梯度洗脫(0-5min,5%A→40%A;5-15 min,40%A→50%A;15-20 min,50%A→63%A;20-35 min,63%A→100%A),流速0.8 ml·min~(-1),330nm(毛蕊花糖苷、蒙花苷),350nm(木犀草素),338nm(芹菜素),柱溫25℃。結(jié)果在所建立色譜條件下各成分的專屬性良好,無干擾峰;線性關(guān)系良好,均在0.9990以上;回收率在93.34%~104.69%,RSD均小于2%,回收率符合要求。測得6產(chǎn)地毛蕊花糖苷、蒙花苷、木犀草素、芹菜素的含量分別為0.3018~1.5269,2.0274~3.2220,0.0536~0.1214,0.3175~0.5590mg/g。結(jié)論經(jīng)方法學(xué)驗證,該法可在同一色譜條件下利用不同檢測窗口實現(xiàn)多類組分及多個成分的同時測定。
