【摘要】：目的:建立并驗證超高效液相色譜法(UPLC)測定人血漿中阿司匹林(ASP)和水楊酸(SA)的濃度,并應(yīng)用于臨床ASP和SA血藥濃度的測定。方法:色譜柱為ACQUITYUPLC~汶BEH C_(18)(2.1 mm×100 mm,1.7μm),流動相為含0.05%三氟乙酸(TFA)的超純水和乙腈(78%∶22%),流速0.5 mL·min~(-1),柱溫30℃,進(jìn)樣器4℃,檢測波長304 nm和277 nm,內(nèi)標(biāo)為替硝唑。結(jié)果:ASP和SA分別在0.4~100μg·mL~(-1)和0.2~50μg·mL~(-1)線性關(guān)系良好(r~20.999);ASP和SA的日內(nèi)、日間準(zhǔn)確度和精密度均15%。結(jié)論:本研究建立了一個準(zhǔn)確、靈敏且穩(wěn)定的UPLC法測定ASP和SA血藥濃度的方法,可應(yīng)用于臨床ASP和SA血藥濃度的測定和藥動學(xué)研究,指導(dǎo)臨床合理用藥。
