基于超高效液相色譜-高分辨質(zhì)譜聯(lián)用技術(shù)的桑黃干預(yù)后大鼠尿液代謝組學(xué)分析

【摘要】:為探討服用桑黃水煎液對機(jī)體的影響,采用超高效液相色譜-高分辨質(zhì)譜(UPLC-HDMS)聯(lián)用技術(shù),檢測灌胃給予桑黃水煎液后大鼠尿液中代謝物的變化。采用正交偏最小二乘法判別分析(OPLS-DA)對空白組和給藥組大鼠尿液代謝物進(jìn)行聚類分析,篩選出潛在的生物標(biāo)志物,并通過MetaboAnalyst 3.0網(wǎng)站分析相關(guān)代謝通路。數(shù)據(jù)顯示,兩組大鼠尿液中的代謝物在第28天得到了很好的區(qū)分,發(fā)現(xiàn)并鑒定了10個生物標(biāo)記物。灌胃給予桑黃水煎液主要對機(jī)體的半胱氨酸和甲硫氨酸代謝、精氨酸和脯氨酸代謝、嘌呤代謝等代謝通路產(chǎn)生影響。研究結(jié)果為深入探討桑黃藥效作用機(jī)制奠定了一定的實驗基礎(chǔ)。
[Abstract]:In order to study the effect of mulberry yellow decoction on the body, the changes of metabolites in urine of rats were detected by ultra-high performance liquid chromatography-high resolution mass spectrometry (UPLC-HDMS). Orthogonal partial least squares discriminant analysis (OPLS-DA) was used to cluster the urine metabolites of rats in blank group and administration group, and potential biomarkers were screened out, and related metabolic pathways were analyzed by MetaboAnalyst 3.0 website. The data showed that the metabolites in the urine of the two groups were well differentiated on the 28th day and 10 biomarkers were found and identified. The administration of decoction mainly affected the metabolism of cysteine and methionine, arginine and proline, and purine metabolism. The results of the study laid a certain experimental foundation for the further study of the mechanism of the effect of mulberry yellow.
