基于UPLC及一測(cè)多評(píng)法同時(shí)測(cè)定蓯蓉總苷膠囊中的活性成分

【摘要】:目的:采用"一測(cè)多評(píng)"(QAMS)技術(shù)建立超高效液相法(UPLC)同時(shí)測(cè)定蓯蓉總苷膠囊中6種成分的分析方法,并與標(biāo)準(zhǔn)曲線法進(jìn)行比較,評(píng)價(jià)一測(cè)多評(píng)法用于蓯蓉總苷膠囊質(zhì)量控制的技術(shù)適應(yīng)性和應(yīng)用可行性。方法:以蓯蓉總苷膠囊為研究對(duì)象,有機(jī)酸類成分以綠原酸為內(nèi)參物,建立其與咖啡酸的相對(duì)校正因子;苯乙醇苷類成分以毛蕊花糖苷為內(nèi)參物,建立其與管花苷A,異毛蕊花糖苷,松果菊苷間的相對(duì)校正因子,同時(shí)考察了各相對(duì)校正因子的耐用性,并比較該方法與標(biāo)準(zhǔn)曲線法測(cè)定結(jié)果的差異,以驗(yàn)證一測(cè)多評(píng)法的準(zhǔn)確性和可行性。結(jié)果:有機(jī)酚酸類成分綠原酸與咖啡酸的相對(duì)校正因子為0.538,苯乙醇苷類成分毛蕊花糖苷與管花苷A,異毛蕊花糖苷,松果菊苷間的相對(duì)校正因子分別為1.346,1.266和1.135,各相對(duì)校正因子的耐用性較好,10批樣品QAMS法的計(jì)算結(jié)果與標(biāo)準(zhǔn)曲線法的測(cè)定結(jié)果無顯著性差異,實(shí)驗(yàn)所得的相對(duì)校正因子可信。結(jié)論:QAMS法可以作為一種簡(jiǎn)便可靠的質(zhì)量評(píng)價(jià)模式用于蓯蓉總苷膠囊多種成分的定量測(cè)定方法。
[Abstract]:Objective: to establish a method for the simultaneous determination of six components in Cistanche glycoside capsules by using "one test and multiple evaluation" QAMS technique, and to compare it with the standard curve method. To evaluate the technical adaptability and application feasibility of one-test multi-evaluation method for quality control of Cistanche glycoside capsules. Methods: taking Cistanche glycoside capsule as the research object, the organic acids were used as the internal reference material, and the relative calibration factor between the organic acids and caffeic acid was established, and the phenylethanoside was used as the internal reference material, and the phenylethanoside was used as the internal reference. The relative correction factors were established and the durability of the relative correction factors were investigated, and the difference between the method and the standard curve method was compared. In order to verify the accuracy and feasibility of a multi-evaluation method. Results: the relative correction factor of Lv Yuan acid and caffeic acid was 0.538. The relative correction factors were 1.346 and 1.135, respectively. The results of QAMS method for 10 batches of samples had no significant difference with that of the standard curve method. The relative correction factors obtained from the experiments were credible. Conclusion this method can be used as a simple and reliable method for the quantitative determination of various components in Cistanche glycoside capsules.
