NHMA改性PNIPAM微凝膠的制備與正溫度響應(yīng)效應(yīng)研究

【摘要】:溫度敏感型聚合物作為有效的藥物載體,可以實現(xiàn)藥物在外界信號作用下的控制釋放。其中聚N-異丙基丙烯酰胺(PNIPAM)是一種較為常見和應(yīng)用較多的溫敏材料,將其制備成為微凝膠形態(tài)并實現(xiàn)正溫度響應(yīng)效應(yīng)的釋放作用,意義明顯。本文用乳液聚合法制備PNIPAM微凝膠,在交聯(lián)劑用量為0.03g、0.05g、0.07g、0.09g、0.11g條件下制備微凝膠試樣,考察交聯(lián)劑用量與PNIPAM微凝膠粒徑的關(guān)系。引入N-羥甲基丙烯酰胺(NHMA)單體,制備共聚的P(NIPAM-co-NHMA)微凝膠,通過設(shè)置交聯(lián)劑用量為0.01g、0.02g、0.03g、0.04g、0.05g和表面活性劑用量為0.01g、0.02g、0.03g、0.05g獲得兩組微凝膠試樣,考察交聯(lián)劑和表面活性劑的用量與P(NIPAM-co-NHMA)微凝膠粒徑的關(guān)系;進一步考察NHMA對共聚微凝膠最低臨界溶解溫度(LCST)的影響,其中NIPAM:NHMA的質(zhì)量比分別為1.5:0、1.4:0.1、1.3:0.2、1.2:0.3、1.1:0.4,結(jié)果表明共聚微凝膠的LCST值調(diào)節(jié)在33℃~40℃之間。采用FT-IR確定了均聚和共聚微凝膠的組成;采用掃描電鏡和透射電鏡觀測均聚和共聚微凝膠的微觀形貌和粒徑。在獲得均聚和共聚微凝膠的基礎(chǔ)上,考察交聯(lián)劑用量、微凝膠溶液濃度、p H值、Na Cl和表面活性劑(SDS)等因素對微凝膠LCST值的影響,結(jié)果表明SDS使微凝膠的LCST值升高,酸和堿使微凝膠的LCST值降低,交聯(lián)劑用量、微凝膠溶液濃度和Na Cl不影響微凝膠的LCST值。以羅丹明B為模型藥物,以雙組份的聚二甲基硅氧烷為成膜基質(zhì),通過流延成膜法制備了負載微凝膠的復合體系,考察了30℃和45℃時薄膜對羅丹明B的釋放量。當溫度高于微凝膠粒子的LCST值時,微凝膠粒子體積會收縮,會有孔隙出現(xiàn),從而羅丹明B通過薄膜的釋放量增加。表明薄膜能夠?qū)崿F(xiàn)正向溫度控制釋放,且隨著薄膜中凝膠粒子復合量的增加,以及隨著薄膜厚度減小,羅丹明B釋放速率相應(yīng)增加。
[Abstract]:As an effective drug carrier, temperature-sensitive polymers can realize controlled release of drugs under the action of external signals. Poly (N- isopropylacrylamide) (PNIPAM) is a common and widely used thermo-sensitive material. It is of great significance to prepare it into microgel form and realize the release of positive temperature response effect. In this paper, PNIPAM microgel was prepared by emulsion polymerization. The microgel samples were prepared under the condition of 0.03g / 0.05g ~ 0.07g ~ 0.07g ~ (0.07g) ~ (0.11) g / g crosslinking agent. The relationship between the amount of crosslinking agent and the particle size of PNIPAM microgel was investigated. The copolymerized PNIPAM-co-NHMA-microgel was prepared by introducing N-hydroxymethylacrylamide (NHMA) monomer. Two groups of microgel samples were obtained by setting the amount of crosslinking agent 0.01g / g (0.02g / g) 0.03g / (0.03g / g) and the amount of surfactant 0.01g / 0.02g / g / 0.03g / 0.05g respectively. The relationship between the amount of crosslinking agent and surfactant and the particle size of PNIPAM-co-NHMA microgel was investigated, and the effect of NHMA on the minimum critical solution temperature (LCST) of copolymer microgel was investigated. The mass ratio of NIPAM: NHMA is 1.5: 0: 1.4: 0.1: 1.3: 1.2: 0.2: 1.1: 0.4. The results show that the LCST value of copolymerized microgel is adjusted between 33 鈩,
