反向電滲流非水毛細(xì)管電泳法快速測定微量赤霉素

【摘要】:赤霉素是一類重要的植物激素,是結(jié)構(gòu)相似的弱酸性二萜類化合物。針對毛細(xì)管電泳法分離、測定赤霉素速度慢、效率不佳等問題,該文發(fā)展了一種可快速測定微量活性赤霉素的非水毛細(xì)管電泳-紫外檢測法。以聚環(huán)氧乙烷動態(tài)涂布毛細(xì)管,利用正離子使電滲流反向,將含10 mmol/L醋酸銨的甲醇-水(95∶5,v/v)作為緩沖體系,在0.08%(v/v)醋酸含量下,分離8種內(nèi)源性赤霉素。結(jié)果表明,赤霉素出峰時間的相對標(biāo)準(zhǔn)偏差(RSD)≤2.1%(日內(nèi))或≤4.3%(日間),峰面積的RSD≤4.5%(日內(nèi))或≤6.9%(日間),檢出限(S/N=3)為1.04~2.20 mg/L,相關(guān)系數(shù)為0.998 2~0.999 3,回收率為87.2%~93.5%。該方法簡單、快速、穩(wěn)定,以含醋酸銨的甲醇水溶液為緩沖體系,預(yù)先考慮了質(zhì)譜測定的需要,可用于實際樣品如麥芽中赤霉素的分析,具有一定的應(yīng)用價值。
[Abstract]:Gibberellin is an important plant hormone and a weak acid diterpenoid with similar structure. Aiming at the problems of separation of gibberellin by capillary electrophoresis, the determination of gibberellin was slow and the efficiency was not good, a non-aqueous capillary electrophoresis and ultraviolet detection method was developed for the rapid determination of trace active gibberellin. The capillary was dynamically coated with poly (ethylene oxide), and the electroosmotic flow was reversed by positive ions. Using methanol-water 95: 5 v / v) containing 10 mmol / L ammonium acetate as buffer system, eight kinds of endogenous gibberellin were separated under the content of 0.08% v / v) acetic acid.
