新型稀土配合物熒光探針的設(shè)計(jì)合成與應(yīng)用

【摘要】:近20年間,以稀土熒光配合物作為探針的時(shí)間分辨熒光測定技術(shù)取得了長足的發(fā)展,并且在生命科學(xué)研究和醫(yī)學(xué)臨床診斷中獲得了越來越多的關(guān)注。同時(shí),單線態(tài)氧(lO2)作為一種重要的活性氧在許多光化學(xué)和光生物化學(xué)反應(yīng)過程中都扮演著重要的角色,特別是在癌癥的光動(dòng)力學(xué)治療技術(shù)(PDT)中單線態(tài)氧起著十分重要的作用。本學(xué)位論文中,.將9,10-二甲基蒽作為單線態(tài)氧識(shí)別基團(tuán),與稀土熒光β二酮銪配合物共價(jià)偶聯(lián),設(shè)計(jì)合成了一種新型的β-二酮銪作為配位體檢測單線態(tài)氧的時(shí)間分辨熒光探針,[Eu(pfdap)3(tpy)] (Hpfdap:1,1,1,2,2-五氟-5-(9',10'-二甲基-2'-乙;)-3,5-乙酰丙酮;tpy:2,2',2”-三聯(lián)吡啶),建立了基于該探針的單線態(tài)氧時(shí)間分辨熒光測定及其在活細(xì)胞內(nèi)檢測單線態(tài)氧的熒光生物成像技術(shù)。由于探針結(jié)構(gòu)中9,10-二甲基蒽基團(tuán)對(duì)銪配合物熒光的淬滅作用,該探針幾乎沒有熒光,而與單線態(tài)氧特異性響應(yīng)之后,9,10-二甲基蒽基團(tuán)被氧化,熒光淬滅作用消失,銪配合物長壽命的熒光得以恢復(fù),其熒光強(qiáng)度增加68倍。該探針選擇性好,靈敏度和信噪比高,適于復(fù)雜生物體系中單線態(tài)氧的測定。利用時(shí)間分辨熒光顯微鏡成像技術(shù),該探針成功用于肝癌細(xì)胞HepG2細(xì)胞在癌癥光動(dòng)力學(xué)治療過程中,光敏化劑5-氨基酮戊酸誘導(dǎo)產(chǎn)生的單線態(tài)氧實(shí)時(shí)原位的示蹤檢測。通過共聚焦顯微鏡和與商業(yè)化線粒體靶向熒光染料共染色實(shí)驗(yàn),進(jìn)一步證明了[Eu(pfdap)3(tpy)]探針的線粒體靶向定位功能,為細(xì)胞器靶向稀土配合物熒光探針的開發(fā)提供了新的思路。
[Abstract]:In recent 20 years, time-resolved fluorimetry with rare earth fluorescent complexes as a probe has made great progress, and has gained more and more attention in life science research and medical clinical diagnosis. At the same time, as a kind of important reactive oxygen species, singlet oxygen species play an important role in many photochemical and photochemical reactions, especially in PDT (photodynamic therapy of cancer). In this thesis. The single line oxygen recognition group of 9 ~ (10) -dimethyl anthracene was covalently coupled with a rare earth fluorescent 尾 -diketone europium complex. A new type of 尾 -dione europium has been designed and synthesized as a time-resolved fluorescence probe for the detection of singlet oxygen. A novel time-resolved fluorescence technique for the detection of singlet oxygen in living cells and a fluorescence biometric imaging technique for the detection of singlet oxygen in living cells have been developed based on tpy: 2 / 2 "- tripyridine. Because the fluorescence of europium complex was quenched by the 9 ~ (10) -dimethyl anthracene group in the structure of the probe, the probe had almost no fluorescence, but the fluorescence quenching effect disappeared after the specific response to the singlet oxygen, and the fluorescence quenching effect disappeared after the oxidation of the 9 ~ (10) -dimethyl anthracene group. The long lifetime fluorescence of europium complex can be recovered and its fluorescence intensity is increased by 68 times. The probe has good selectivity, high sensitivity and signal-to-noise ratio, and is suitable for the determination of singlet oxygen in complex biological systems. Using time-resolved fluorescence microscopy, the probe was successfully used to detect in situ single linear oxygen produced by 5-aminolevulinic acid (5-aminolevulinic acid) during photodynamic therapy of HepG2 cells. By means of confocal microscopy and co-staining with commercial mitochondria targeted fluorescent dyes, the mitochondrial targeting localization function of Eupfdapia 3tpy3 probe is further proved, which provides a new idea for the development of organelle targeting rare earth complex fluorescent probes.
