用于檢測次氯酸根和汞離子的熒光探針的設(shè)計(jì)合成及應(yīng)用

【摘要】:熒光分析法操作簡單,靈敏度高,能實(shí)現(xiàn)生物體內(nèi)的實(shí)時(shí)動態(tài)監(jiān)測,在醫(yī)學(xué)、生命科學(xué)以及環(huán)境科學(xué)等領(lǐng)域得到廣泛的應(yīng)用。本論文正是基于熒光檢測法設(shè)計(jì)合成了三種分別檢測HClO和Hg~(2+)的熒光探針:1.我們設(shè)計(jì)合成一種基于2-(2'-羥基苯基)苯并噻唑(HBT)的新型“turn-on”型Cl O-熒光探針PHC2;贑lO~-選擇性氧化斷裂偶氮鍵,探針對ClO~-顯示出高度特異性選擇性和高靈敏度。2.我們合成了疊氮基取代的2-(2'-羥基苯基)苯并噻唑(HBT)的“turn-on”型ClO~-熒光探針PHC3。探針在PBS:EtOH(V/V=8:2,pH 7.4,1%MeCN)的體系下對ClO~-有著良好特異性和靈敏度。3.我們以香豆素為熒光團(tuán),硫縮醛為報(bào)告基團(tuán)而設(shè)計(jì)合成了“比率計(jì)”型熒光響應(yīng)的汞離子探針PHC4。在PBS(pH 7.4)的檢測體系下,體系呈現(xiàn)藍(lán)色熒光,而隨著Hg~(2+)的加入,體系在497 nm處的藍(lán)色熒光峰逐漸降低,而在566 nm處產(chǎn)生新的黃色熒光峰,并不斷增強(qiáng),形成明顯的比率計(jì)型熒光響應(yīng)。此外,探針還實(shí)現(xiàn)了在海拉細(xì)胞中汞離子的熒光成像。
[Abstract]:Fluorescence analysis is simple in operation and high in sensitivity. It can be used in real-time dynamic monitoring of organisms and has been widely used in medicine, life science and environmental science. In this paper, we have designed and synthesized three fluorescent probes for detecting HClO and Hgnion 2, respectively, based on the fluorescence detection method. We have designed and synthesized a novel "turn-on" type ClO- fluorescence probe PHC _ 2 based on 2-butadiene-hydroxyphenyl) benzothiazolium (HBT). Based on the cleavage of azo bond by selective oxidation, the probe showed high selectivity and sensitivity to ClO-. We have synthesized the "turn-on" type ClOI-fluorescence probe PHC3 of azido-substituted 2-oxo-hydroxyphenyl) benzothiazolium (HBT). The probe has good specificity and sensitivity to ClO- in the system of PBS: EtOHN / V / V = 8: 2, pH 7.4 ~ (-1) MeCN-1). Using coumarin as the fluorescence group and thioacetal as the reporting group, we have designed and synthesized a "ratio" type mercury ion probe PHC4 with fluorescence response. The blue fluorescence was observed in the system of PBSX pH 7.4). The blue fluorescence peak at 497 nm decreased and the yellow fluorescence peak was increased at 566nm with the addition of Hgf2). An obvious ratio of fluorescence response is formed. In addition, the probe also realized the fluorescence imaging of mercury ions in Haila cells.
