碩樺化學(xué)成分的研究

【摘要】:碩樺(Betula costata Trautv.)為樺木科樺木屬的植物,其樹(shù)皮具有清熱解毒、止咳、平喘、消炎、殺菌等功效,用于治療慢性痢疾、血尿以及粉刺、雀斑等疾病。本文從碩樺樹(shù)皮的85%乙醇提取物中分離得到9個(gè)單體化合物(分別標(biāo)記為SH-1~SH-9),經(jīng)核磁、紅外光譜等數(shù)據(jù)分析,結(jié)合理化性質(zhì),鑒定它們分別是白樺脂醇(SH-1),羽扇豆醇(SH-2),3-O-乙酰齊墩果酸(SH-3),樺木酮酸(SH-4),樺木酮(SH-5),17α-Hydroxy-28-nor-18α(H)-lup-20(29)-en-3-one(SH-6),苦杏仁苷(SH-7),槭木素E(SH-8)和16-羥基-17-甲氧基-槭木素E(SH-9)。其中,17α-Hydroxy-28-nor-18α(H)-lup-20(29)-en-3-one為首次從天然產(chǎn)物中分離得到的化合物,16-羥基-17-甲氧基-槭木素E和苦杏仁苷為首次從碩樺中分離得到的化合物。在化學(xué)成分的研究基礎(chǔ)上,本論文首次建立了兩種三萜類(lèi)化合物碩樺樹(shù)皮中羽扇豆醇(化合物SH-2)和3-O-乙酰齊墩果酸(化合物SH-3)含量的HPLC測(cè)定方法。首先,對(duì)提取溶劑、提取方法、提取時(shí)間以及液料比等影響因素進(jìn)行了考察,確立了最佳的提取方法。其次,進(jìn)行了方法學(xué)考察。實(shí)驗(yàn)結(jié)果表明,化合物SH-2和SH-3分別在0.08-2.50 mg/mL和0.16-5.00 mg/mL范圍內(nèi)峰面積(Y)和進(jìn)樣濃度(X)呈良好的線性關(guān)系,化合物SH-2和SH-3分析方法的日內(nèi)精密度、日間精密度、重復(fù)性、穩(wěn)定性、加樣回收率的RSD均小于2%,說(shuō)明所建立的含量測(cè)定方法具有精密度好、準(zhǔn)確度高等優(yōu)點(diǎn)。含量測(cè)定結(jié)果,采自新賓縣的碩樺中SH-2的含量為1.83 mg/g,SH-3含量為5.25 mg/g,采自靖宇縣的碩樺中SH-2的含量為0.14 mg/g,SH-3含量為4.66mg/g,采自靖宇縣的白樺SH-2的含量為1.15 mg/g,SH-3含量為1.25 mg/g。表明不僅同屬植物碩樺和白樺之間SH-2、SH-3化合物含量差異較大,而且不同產(chǎn)地的碩樺之間含量差別也很大。綜上,在國(guó)內(nèi)外學(xué)者研究的基礎(chǔ)上,本文從碩樺皮乙醇提取物中共計(jì)分離鑒定了9個(gè)化合物,其中有一個(gè)為首次從天然產(chǎn)物中分離得到的化合物,2個(gè)為首次從碩樺中分得的化合物;同時(shí)還建立了碩樺皮中兩種五環(huán)三萜類(lèi)化合物的HPLC-DAD含量測(cè)定方法。本文的研究結(jié)果為建立碩樺的質(zhì)量標(biāo)準(zhǔn)以及碩樺的開(kāi)發(fā)利用提供了新的科學(xué)依據(jù)。
[Abstract]:Betula costata Trautv. It is a kind of birch plant in the family Betulaceae. Its bark is used to treat chronic dysentery, hematuria, acne, freckle and other diseases with antipyretic, antitussive, antiasthmatic, anti-inflammatory and bactericidal effects. In this paper, nine monomers (labeled as SH-1 and SH-9) were isolated from 85% ethanol extract from the bark of birch bark. The compounds were analyzed by NMR, IR and physicochemical properties. They were identified as Bai Hua aliphatic alcohol (SH-1), Lupanolol (SH-2) -acetyl oleanolic acid (SH-3), betulinic acid (SH-4), betulinone (17 偽 -Hydroxy-28-nor-18 偽) H-lup-2029-en-3-one (SH-6), amygdalin (SH-7), maple (ESH-8) and 16-hydroxy-17-methoxy (ESH-9). Among these compounds, 17 偽 -Hydroxy-28-nor-18 偽 -Han-lup-2029-en-3-one was isolated from natural products for the first time, and the compounds of 16-hydroxy-17-methoxy-maple lignin E and amygdalin were first isolated from birch. Based on the study of chemical constituents, a HPLC method for the determination of the contents of lupinol (SH-2) and 3-O- acetyl oleanolic acid (compound SH-3) in the bark of two triterpenoids was established for the first time. Firstly, the influence factors such as solvent, extraction method, extraction time and the ratio of liquid to material were investigated, and the best extraction method was established. Secondly, the methodology is investigated. The experimental results showed that the peak area of SH-2 and SH-3 were linear in the range of 0.08-2.50 mg/mL and 0.16-5.00 mg/mL, respectively. The precision, repeatability and stability of compound SH-2 and SH-3 were obtained. The RSD of the recoveries was less than 2, which indicated that the established method had the advantages of good precision and high accuracy. The results showed that the content of SH-2 was 1.83 mg / g SH-3 was 5.25 mg / g, the content of SH-2 was 0.14 mg / g SH-3 was 4.66 mg / g, the content of Bai Hua SH-2 was 1.15 mg / g SH-3 in Jingyu county. The results showed that not only the content of SH-2N SH-3 was different between the same genus Betula platyphylla and Bai Hua, but also the content of SH-2SH-3 was different among different habitats. In summary, based on the research of domestic and foreign scholars, nine compounds were isolated and identified from the ethanol extracts of Betula platyphylla. One of the compounds was isolated from natural products for the first time and two were isolated from Betula platyphylla for the first time. A HPLC-DAD method for the determination of two pentacyclic triterpenoids in Betula platyphylla bark was also established. The results of this paper provide a new scientific basis for the establishment of quality standards and the development and utilization of birch.
