苦丁茶冬青胰脂肪酶抑制活性成分研究

【摘要】:本論文通過體外活性篩選,發(fā)現(xiàn)了苦丁茶冬青(Ilex kudingcha C.J.Tseng)的甲醇提取物具有胰脂肪酶抑制活性,然后以胰脂肪酶抑制活性篩選為指導(dǎo),對苦丁茶冬青的醇提取物進(jìn)行活性追蹤導(dǎo)向分離,得到了苦丁茶冬青中的活性部位和主要活性成分,再采用LC-MS方法對活性部位的化學(xué)組成進(jìn)行了闡述,并對主要活性成分對胰脂肪酶的抑制行為進(jìn)行了初步研究。主要內(nèi)容如下:(1)以胰脂肪酶抑制活性篩選為指導(dǎo),發(fā)現(xiàn)了苦丁茶冬青提取物的活性部位,利用多種色譜手段對活性部位進(jìn)行分離,得到5個化合物(1-5),經(jīng)1H-NMR、13C-NMR、HR-ESI-MS等方法,結(jié)合文獻(xiàn)數(shù)據(jù),分別鑒定為:3-O-[?-L-rhamnopyranosyl(1-2)]-[?-D-glucopyranosyl(1 3)-]-?-L-arabinopyranosyl pomolic acid 28-O-[?-Lrhamnopyranosyl(1 2)]-?-D-glucopyranoside(1),3-O-[?-L-rhamnopyranosyl(1 2)]-[?-D-glucopyranosyl(1 3)-]-?-L-arabino-pyranosyl siaresinolic acid28-O-[?-L-rhamnopyranosyl(1 2)]-?-D-glucopyranoside(2),3-O-?-D-glucopyranosyl(1-2)?-D(1-2)-?-D-glucopyranosyl(1-3)-[?-L-rhamnopyranosyl(1-2)]?-L-arabinopyranosyl pomolic acid28-O-?-L-rhamnopyranosyl(1-2)?-D-glucopyranoside(3),3-O-?-D-glucopyranosyl(1-2)-?-D-glucopyranosyl(1-3)-[?-L-rhamnopyranosy l(1-2)]-?-L-arabinopyanosyl siaresinolic acid 28-O-?-L-rhamnopyranosyl(1-2)?-D-glucopyranoside(4),3-O-?-L-rhamnopyranosyl(1-2)-[?-Dglucopyranosyl(1 3)]-?-L-arabinopyranosyl kudinolic acid 28-O-?-Lrhamnopyranosyl(1 2)-?-D-glucopyranoside(5)。(2)測定了分離得到的活性化合物1-5對抑制脂肪酶活性的IC50以及這5個化合物單體對胰脂肪酶活性的抑制類型。它們的IC50分別是13.2、13.0、38.1、28.7、22.2μM,對胰脂肪酶的抑制均為非競爭性抑制。(3)研究了化合物1-5的質(zhì)譜裂解規(guī)律,歸納出了各個化合物質(zhì)譜碎片峰裂解途徑,并在此基礎(chǔ)上,利用UPLC-Q-TOF-HRESI-MS方法,對苦丁茶冬青胰脂肪酶抑制活性部位的化學(xué)成分進(jìn)行了歸屬,給出了活性部位的化學(xué)特征。結(jié)果說明苦丁茶冬青活性部位的化學(xué)成分由三萜皂苷組成,其中主要組分Latifolioside G,H和Kudinoside N,O和I五個組分的含量,超過總苷的85%。其他組分分子結(jié)構(gòu)類似,也說明了該類三萜皂苷結(jié)構(gòu)的細(xì)微變化對胰脂肪酶抑制活性影響不大。
[Abstract]:In this paper, we found that the methanol extract of Ilex kudingcha C.J.Tseng) had the inhibitory activity of pancreatic lipase, and was guided by the screening of lipase inhibitory activity of Pancreatic lipase. The active parts and main active components of Ilex kudingcha were obtained by activity tracing and guiding separation. The chemical composition of the active parts was described by LC-MS method. The inhibitory action of main active components on pancreatic lipase was studied. The main contents are as follows: (1) guided by the screening of lipase inhibitory activity, the active site of the extract of Ilex kudingcha was found. The active parts were separated by a variety of chromatographic methods, and five compounds were obtained by 1H-NMR-13C-NMR-HR-ESI-MS. Combined with literature data, 鍒嗗埆閴村畾涓,
