復(fù)方板藍(lán)根顆粒APCI-MS指紋圖譜及化學(xué)成分的LC-APCI-MS~n研究

【摘要】:建立了復(fù)方板藍(lán)根顆粒提取物的大氣壓化學(xué)電離質(zhì)譜(APCI-MS)的指紋圖譜分析方法,并利用液相色譜-大氣壓化學(xué)電離-串聯(lián)質(zhì)譜(LC-APCI-MS~n)技術(shù)對(duì)主要指紋峰的歸屬進(jìn)行確認(rèn)。結(jié)果表明:質(zhì)譜指紋圖譜確定了10批不同來(lái)源復(fù)方板藍(lán)根顆粒提取物的10個(gè)共有峰,各提取物的指紋圖譜相似度均在97%以上。化學(xué)成分的LC-APCI-MS~n分析鑒別了復(fù)方板藍(lán)根顆粒提取物中吲哚類、奎唑酮類、有機(jī)酸類和卟啉類等12個(gè)化合物,并確定了5個(gè)質(zhì)譜指紋峰的歸屬。采用體外實(shí)驗(yàn)研究對(duì)復(fù)方板藍(lán)根顆粒提取物的抗病毒活性進(jìn)行評(píng)價(jià),結(jié)果表明,其具有一定的抗病毒活性。該指紋圖譜的精密度、穩(wěn)定性和重現(xiàn)性良好,化學(xué)成分分析快捷、靈敏,結(jié)合化學(xué)成分分析和藥效學(xué)研究,可為復(fù)方板藍(lán)根顆粒質(zhì)量控制標(biāo)準(zhǔn)的制訂奠定基礎(chǔ),同時(shí)也為深入研究其藥效物質(zhì)基礎(chǔ)和譜效關(guān)系提供理論依據(jù)。
[Abstract]:A fingerprint analysis method for APCI-MS of compound Isatis root granule extract was established. The main fingerprint peaks were confirmed by liquid chromatography-atmospheric pressure chemical ionization tandem mass spectrometry (LC-APCI-MSN) technique. The results showed that the 10 common peaks of 10 batches of compound Radix Isatidis granules extracts were identified by mass spectrometry, and the fingerprint similarity of each extract was more than 97%. LC-APCI-MS~n analysis of the chemical constituents identified 12 compounds such as indole, quezolidone, organic acids and porphyrins in compound Isatis root granule extracts, and determined the attribution of five MS fingerprint peaks. The antiviral activity of compound Radix Isatidis granules extract was evaluated in vitro. The results showed that it had certain antiviral activity. The fingerprint has good precision, stability and reproducibility, quick and sensitive chemical component analysis, combined with chemical composition analysis and pharmacodynamics study, can lay a foundation for the formulation of quality control standard of compound Isatis root granules. At the same time, it also provides the theoretical basis for the further study of the substance basis and spectrum effect relationship of its pharmacodynamics.
