MALDI-TOF MS磷脂質(zhì)組學(xué)快速分析三文魚肌肉組織

【摘要】:建立了基質(zhì)輔助激光解吸電離-飛行時間質(zhì)譜(MALDI-TOF MS)快速分析三文魚肌肉組織磷脂質(zhì)組學(xué)的方法。以非自然磷脂酰膽堿DMPC(14∶0/14∶0)和磷脂酰乙醇胺DPPE(15∶0/15∶0)為標(biāo)準(zhǔn)品,分析其在正離子模式下的主要分子峰與干擾峰,并進行方法學(xué)驗證。實驗結(jié)果表明,各化合物平均加標(biāo)回收率在74%~83%之間,相對標(biāo)準(zhǔn)偏差小于7%,日內(nèi)和日間精密度的相對標(biāo)準(zhǔn)偏差均低于8.5%。將三文魚磷脂提取物進樣分析,共鑒定出28種磷脂分子,其中m/z806.40([PC38:6+H]~+和[PE38:4+K]~+)信號最強。三文魚中多不飽和磷脂種類豐富,如m/z 824.38([PC38:8+Na]~+和[PE42:5+H]~+),m/z832.41([PC40:7+H]~+,[PC38:4+Na]~+和[PE40:5+H]~+),部分磷脂分子的脂肪酸鏈達到了滿不飽和,如[PC42:11+H]~+,其sn-1和sn-2上兩條脂肪酸鏈分別為二十碳五烯酸鏈(EPA-)和二十二碳六烯酸鏈(DHA-)。該方法穩(wěn)定可靠,可用于鑒定三文魚肌肉組織中的磷脂分子結(jié)構(gòu),并從脂質(zhì)組學(xué)角度為三文魚營養(yǎng)評價提供理論依據(jù)。
[Abstract]:A method for rapid analysis of phospholipid in salmon muscle tissue by matrix assisted laser desorption ionization and time of flight mass spectrometry (MALDI-TOF) was established. Unnatural phosphatidylcholine (DMPC) (14: 0: 14: 0) and phosphatidylethanolamine (DPPE15: 0 / 15: 0) were used as standard materials. The main molecular peaks and interference peaks in positive ion mode were analyzed and the methodology was verified. The results showed that the average recoveries of each compound were between 74 and 83%, the relative standard deviation was less than 7, and the relative standard deviation of intra-day and inter-day precision was lower than 8.5%. A total of 28 phospholipid molecules were identified by injection analysis of phospholipid extracts from salmon, of which m / z 806.40 ([PC38:6 H] ~ and [PE38:4 K] ~) signals were the strongest. The species of polyunsaturated phospholipid in salmon are rich, such as m / z 824.38 ([PC38:8 Na] ~ and [PE42:5 H] ~ + 832.41 ([PC40:7 H] ~, [PC38:4 Na] ~ and [PE40:5 H] ~). Some of the fatty acid chains of phospholipid molecules have reached full unsaturated, such as [PC42:11 H] ~, The two fatty acid chains on sn-1 and sn-2 are eicosapentaenoic acid chain EPA-) and 22 carbohexaenoic acid chain DHA-C, respectively. The method is stable and reliable and can be used to identify the molecular structure of phospholipid in salmon muscle tissue and provide theoretical basis for nutrition evaluation of salmon from the point of view of lipid group.
