天然產(chǎn)物來(lái)源多糖結(jié)構(gòu)解析研究進(jìn)展

【摘要】:多糖是高等植物、動(dòng)物、微生物細(xì)胞中一類非常重要的天然大分子物質(zhì),由于許多活性多糖具有良好的免疫調(diào)節(jié)、抗腫瘤、抗氧化和調(diào)節(jié)腸道菌群等功能,因此科學(xué)認(rèn)識(shí)多糖結(jié)構(gòu)有利于深入研究其生物活性功能,對(duì)多糖的基礎(chǔ)研究和應(yīng)用具有重要意義。本文簡(jiǎn)述天然產(chǎn)物來(lái)源多糖分離純化和結(jié)構(gòu)解析的研究方法,重點(diǎn)介紹葡聚糖、葡甘聚糖、木葡聚糖、阿拉伯木聚糖和果膠等多糖結(jié)構(gòu)解析的研究進(jìn)展,就多糖結(jié)構(gòu)解析目前存在的一些問(wèn)題進(jìn)行探討與展望。通過(guò)歸納文獻(xiàn)資料并結(jié)合筆者團(tuán)隊(duì)近20年的研究積累,提出"部分降解-甲基化-核磁共振"可作為多糖一級(jí)結(jié)構(gòu)解析的核心技術(shù),同時(shí)可輔助利用ESI-MS、Maldi-Tof-MS、離子淌度-質(zhì)譜聯(lián)用等技術(shù)與方法,從而構(gòu)建一套先進(jìn)的多糖一級(jí)結(jié)構(gòu)解析技術(shù)體系。
[Abstract]:Polysaccharide is a kind of very important natural macromolecular substance in higher plants, animals and microorganism cells, because many active polysaccharides have good functions of immune regulation, anti-tumor, anti-oxidation and regulation of intestinal flora, etc. Therefore, scientific understanding of the structure of polysaccharides is beneficial to the further study of their biological activity and function, and is of great significance to the basic research and application of polysaccharides. In this paper, the methods of isolation, purification and structure analysis of polysaccharides from natural products are reviewed, and the progress in structural analysis of polysaccharides, such as dextran, glucomannan, xyloglucan, arabinoxylan and pectin, is emphasized. Some problems existing in the structure analysis of polysaccharides were discussed and prospected. By summarizing the literature and combining the research and accumulation of the author's team in the past 20 years, it is proposed that "partial degradation-methylation-nuclear magnetic resonance" can be used as the core technology for the analysis of the primary structure of polysaccharides. At the same time, we can use ESI-MSN Maldi-Tof-MSand ion mobility mass spectrometry to construct a set of advanced primary structure analysis technology system of polysaccharides.
