疏水改性海藻酸鈉的載藥釋藥性能及其與殼聚糖的協(xié)同絮凝作用

【摘要】:海藻酸鈉是一種天然高分子多糖,在自然界中廣泛存在,具有良好的生物相容性、非免疫性、安全性及生物降解性,被廣泛應(yīng)用于生物醫(yī)學(xué)等領(lǐng)域。本文以十二烷基縮水甘油醚為接枝原料對(duì)海藻酸鈉進(jìn)行疏水改性,得到了兩親性的海藻酸鈉接枝產(chǎn)物。通過(guò)表面張力、動(dòng)態(tài)光散射、Zeta電位等研究方法,系統(tǒng)研究了疏水改性海藻酸鈉(HM-alginate)在水溶液中的膠束化行為,以及HM-alginate對(duì)疏水性物質(zhì)的增溶能力和對(duì)藥物的緩釋性能,并對(duì)HM-alginate與殼聚糖的協(xié)同絮凝作用進(jìn)行了研究。1.在堿性條件下,以十二烷基縮水甘油醚為疏水性接枝原料,對(duì)海藻酸鈉進(jìn)行疏水改性,得到了具有兩親性的海藻酸鈉衍生物。所合成的疏水改性海藻酸鈉具有明顯的表面活性,在水溶液中可以形成以疏水性烷氧基鏈為內(nèi)核和以親水性海藻酸鈉為外殼的核-殼結(jié)構(gòu)的膠束,其臨界膠束濃度為0.4 mg/m L。形成膠束時(shí)溶液的粒徑在1000 nm左右,Zeta電位為-63±1 mV,該膠束在溶液中能夠穩(wěn)定存在。2.疏水改性海藻酸鈉對(duì)脂溶性染料蘇丹紅Ⅳ有良好的增溶作用,通過(guò)將蘇丹紅Ⅳ包裹在膠束的疏水內(nèi)核中增加其在溶液中的溶解度。3.疏水改性海藻酸鈉對(duì)疏水藥物氯法齊明和兩性霉素B均有良好的增溶作用,可以使這兩種藥物在溶液中的溶解度分別增大1100倍和160倍。氯法齊明和兩性霉素B在HM-alginate溶液中的溶解度隨NaCl濃度的升高而降低,隨溶液pH的增大而降低。4.疏水改性海藻酸鈉對(duì)氯法齊明及兩性霉素B均有較好的緩釋作用,HM-alginate溶液濃度越大,緩釋效果越明顯。5.在疏水改性海藻酸鈉溶液中加入殼聚糖后可以觀察到明顯的絮凝現(xiàn)象,疏水改性海藻酸鈉與殼聚糖(HM-alginate/chitosan)的絮凝作用可以用來(lái)提取水中的疏水性染料蘇丹紅Ⅳ。HM-alginate/chitosan對(duì)蘇丹紅Ⅳ的提取率隨HM-alginate濃度的增加而降低,隨殼聚糖濃度的增加先升高后降低。pH為4.0時(shí),1.0 mg/m L的HM-alginate和3.0 mg/mL的殼聚糖溶液混合后,對(duì)蘇丹紅Ⅳ的提取效果最好,提取率約為45.1%。6.疏水改性海藻酸鈉與殼聚糖的絮凝作用受溶液pH值的影響較大,在pH值為3.0-3.8時(shí),HM-alginate/chitosan對(duì)蘇丹紅Ⅳ的提取效果最好,提取率在75%左右。7.疏水改性海藻酸鈉與殼聚糖的絮凝作用具有選擇性,可以選擇性除去溶液中的疏水染料蘇丹紅Ⅳ,而對(duì)親水染料亞甲基藍(lán)沒(méi)有顯著影響,且HM-alginate/chitosan對(duì)蘇丹紅Ⅳ的提取率不受溶液中亞甲基藍(lán)存在的影響。在亞甲基藍(lán)存在的條件下,pH值為3.0-3.8時(shí)HM-alginate/chitosan對(duì)蘇丹紅Ⅳ的提取率仍為75%左右,對(duì)亞甲基藍(lán)的提取率在10%以下。
[Abstract]:Sodium alginate is a kind of natural polymer polysaccharide, which is widely existed in nature and has good biocompatibility, non-immunity, safety and biodegradability. It is widely used in biomedicine and other fields. The hydrophobic modification of sodium alginate with dodecyl glycidyl ether was carried out and the amphiphilic sodium alginate grafted product was obtained. The micellization behavior of hydrophobic modified sodium alginate (HM-alginate) in aqueous solution, the solubilization ability of HM-alginate to hydrophobic substances and the sustained release properties of drugs were systematically studied by means of surface tension, dynamic light scattering and Zeta potential. The synergistic flocculation of HM-alginate and chitosan was studied. The hydrophobic modification of sodium alginate with dodecyl glycidyl ether as hydrophobic graft material was carried out under alkaline conditions, and a amphiphilic sodium alginate derivative was obtained. The hydrophobic modified sodium alginate has obvious surface activity and can form core-shell micelles with hydrophobic alkoxyl chain as core and hydrophilic sodium alginate as shell in aqueous solution. The critical micelle concentration is 0.4 mg/m / L. The particle size of the solution was about 1000 nm and the Zeta potential was -63 鹵1 MV. The micelle could exist stably in the solution. The hydrophobic modified sodium alginate has a good solubilization effect on the liposoluble dye Sudan 鈪,
