谷胱甘肽熒光探針的合成及性質(zhì)研究

【摘要】:谷胱甘肽是生物體內(nèi)重要的活性物質(zhì),具有許多生理功能。研究已表明谷胱甘肽可用于體內(nèi)氟化物、丙烯腈、重金屬及有機(jī)溶劑等的中毒治療;保護(hù)細(xì)胞膜免受氧化性破壞;促進(jìn)高鐵血紅蛋白的還原;保護(hù)由腫瘤藥物、放射性藥物等引起的白細(xì)胞減少癥狀;緩解由缺氧血癥、惡心以及肝臟疾病所引起的不適等功能。然而體內(nèi)谷胱甘肽含量過(guò)高或過(guò)低都會(huì)影響正常的生理機(jī)能,導(dǎo)致一些疾病的產(chǎn)生如肝損傷、癌癥、艾滋病、骨質(zhì)疏松癥、阿耳茨海默氏病、心血管疾病等,所以維持體內(nèi)谷胱甘肽含量的穩(wěn)定是非常重要的。由于半胱氨酸、同型半胱氨酸與谷胱甘肽具有相似的分子骨架和反應(yīng)活性(如巰基),導(dǎo)致在細(xì)胞內(nèi)高選擇性的檢測(cè)谷胱甘肽相當(dāng)困難。因此,如何快速、高效、準(zhǔn)確和專一性地檢測(cè)細(xì)胞內(nèi)谷胱甘肽具有很大的挑戰(zhàn)。本論文設(shè)計(jì)并合成了三類谷胱甘肽熒光探針。具體工作內(nèi)容如下:1.設(shè)計(jì)并合成了以丹磺酰胺為熒光團(tuán)的銅離子/谷胱甘肽熒光探針。實(shí)驗(yàn)結(jié)果表明,該探針能夠高選擇性地識(shí)別銅離子并不受其它金屬離子的干擾,此外生成的銅離子配合物可進(jìn)一步用于谷胱甘肽的檢測(cè)。2.設(shè)計(jì)并合成了以七甲川菁染料為骨架的谷胱甘肽熒光探針。實(shí)驗(yàn)結(jié)果表明,該探針在生理?xiàng)l件下展現(xiàn)了對(duì)谷胱甘肽的高選擇性并不受其它氨基酸的干擾。細(xì)胞實(shí)驗(yàn)表明,該探針適用于細(xì)胞內(nèi)檢測(cè)谷胱甘肽,并可區(qū)分半胱氨酸和同型半胱氨酸。老鼠成像表明,該探針可運(yùn)用于老鼠體內(nèi)檢測(cè)谷胱甘肽并且產(chǎn)物可以進(jìn)一步代謝排出。3.設(shè)計(jì)并合成了以1,8-萘酰亞胺磺酰胺為骨架的谷胱甘肽熒光探針。熒光光譜數(shù)據(jù)表明,該探針在生理?xiàng)l件下能夠高選擇性地識(shí)別谷胱甘肽并區(qū)分半胱氨酸和同型半胱氨酸。細(xì)胞成像表明,探針可適用于細(xì)胞內(nèi)專一性檢測(cè)谷胱甘肽,并定位于亞細(xì)胞溶酶體內(nèi)檢測(cè)谷胱甘肽。
[Abstract]:Glutathione is an important active substance in organism and has many physiological functions. Studies have shown that glutathione can be used to treat fluoride, acrylonitrile, heavy metals and organic solvents in vivo; to protect cell membranes from oxidative damage; to promote methemoglobin reduction; and to protect against cancer drugs. Leucopenia caused by radiopharmaceuticals; relief from anoxia, nausea, and malaise caused by liver disease. However, high or low levels of glutathione in the body can affect normal physiological function, leading to some diseases such as liver injury, cancer, AIDS, osteoporosis, Alzheimer's disease, cardiovascular disease, etc. Therefore, it is very important to maintain the stability of glutathione content in the body. Because of cysteine, homocysteine and glutathione have similar molecular skeleton and reactive activity (such as thiol), which makes it very difficult to detect glutathione in cells with high selectivity. Therefore, how to detect intracellular glutathione quickly, efficiently, accurately and specifically is a challenge. In this paper, three kinds of glutathione fluorescent probes were designed and synthesized. The details of the work are as follows: 1. Copper ion / glutathione fluorescent probe with dansulfonamide as fluorescence group was designed and synthesized. The experimental results show that the probe can highly selectively recognize copper ions without interference from other metal ions, and the resulting copper ion complexes can be further used for the detection of glutathione. 2. A glutathione fluorescence probe with heptomethylcyanine dyes as skeleton was designed and synthesized. The results showed that the probe exhibited high selectivity to glutathione under physiological conditions without interference by other amino acids. Cell experiments showed that the probe was suitable for intracellular detection of glutathione and could distinguish cysteine from homocysteine. Mouse imaging showed that the probe could be used to detect glutathione in mice and that the product could be metabolized further. A glutathione fluorescence probe was designed and synthesized with 1 D 8 naphthalimide sulfonamide as the skeleton. Fluorescence spectra showed that the probe could selectively recognize glutathione and distinguish cysteine from homocysteine under physiological conditions. Cell imaging showed that the probe could be used to detect glutathione specifically in cells and to detect glutathione in subcellular lysosomes.
