居間駒形氏桿菌發(fā)酵大豆糖蜜生產(chǎn)細(xì)菌纖維素條件的優(yōu)化

【摘要】:通過(guò)居間駒形氏桿菌(Komagataeibacter intermedius CGMCC12562)發(fā)酵生產(chǎn)細(xì)菌纖維素,以大豆糖蜜作主要發(fā)酵原料,優(yōu)化發(fā)酵工藝參數(shù),提高細(xì)菌纖維素產(chǎn)量。在單因素試驗(yàn)的基礎(chǔ)上,篩選出對(duì)細(xì)菌纖維素產(chǎn)量影響較大的4個(gè)因素,即大豆糖蜜可溶性固形物含量、玉米蛋白粉、ZnSO_4及蘋果酸添加量,并通過(guò)Design-Expert響應(yīng)面分析對(duì)這4個(gè)因素進(jìn)行優(yōu)化,得到最優(yōu)的培養(yǎng)基及發(fā)酵工藝為:大豆糖蜜可溶性固形物含量14.13°Brix、玉米蛋白粉添加量1.6%、ZnSO_4添加量0.11%、蘋果酸添加量0.41%;在30℃、接種量10%、裝料量16%、初始pH 6.0的條件下靜置培養(yǎng)7d,細(xì)菌纖維素產(chǎn)量高達(dá)(15.68±0.82)g/L。
[Abstract]:Bacterial cellulose was produced by the fermentation of Komagataeibacter intermedius CGMCC12562. Soybean molasses was used as the main fermentation material to optimize the fermentation parameters and increase the production of bacterial cellulose. On the basis of single factor experiment, four factors were selected to influence the yield of bacterial cellulose, namely, the content of soluble solids in soybean molasses, the content of ZnSO4 in corn protein powder and the amount of malic acid added. The four factors were optimized by Design-Expert response surface analysis. The optimum medium and fermentation technology were obtained as follows: the content of soluble solids in molasses of soybean was 14.13 擄Brix, the amount of corn protein powder was 1.6 and the amount of ZnSO4 was 0.11 and the amount of malic acid was 0.41 at 30 鈩,
