檢測(cè)二氧化硫衍生物、次氯酸根的熒光探針的設(shè)計(jì)合成及生物成像應(yīng)用

【摘要】:二氧化硫衍生物常被做為食物防腐劑而使用在日常生活當(dāng)中,用來(lái)防止食物氧化和細(xì)菌生長(zhǎng)。隨著工業(yè)化的進(jìn)展,促使煤、石油以及化石燃料的大量使用,導(dǎo)致了SO2被大量排放到空氣中。低濃度的二氧化硫衍生物對(duì)人體影響不大,然而,當(dāng)二氧化硫衍生物的濃度超過(guò)一定范圍,便會(huì)對(duì)人體健康產(chǎn)生一定損害,如:呼吸系統(tǒng)疾病、癌癥等。次氯酸是人體在自然氧化代謝過(guò)程中產(chǎn)生的一種副產(chǎn)物,次氯酸與生物體的各項(xiàng)生理和病理過(guò)程有著密切的關(guān)系。當(dāng)生物體內(nèi)次氯酸濃度過(guò)高就會(huì)引起癌癥、關(guān)節(jié)炎等疾病。因此,利用熒光探針技術(shù)定量檢測(cè)二氧化硫衍生物和次氯酸根具有重要的意義。本論文設(shè)計(jì)和合成了用于檢測(cè)亞硫酸根(T1和T2)、亞硫酸氫根(T3)和次氯酸根(T4)的熒光探針,并對(duì)它們的光物理性質(zhì)和生物學(xué)應(yīng)用進(jìn)行了分析研究。研究發(fā)現(xiàn),這幾種探針都具有優(yōu)異的選擇性能,以及良好的光譜信號(hào)響應(yīng)變化,這些探針被成功的應(yīng)用于檢測(cè)活體細(xì)胞和秀麗線(xiàn)蟲(chóng)中的目標(biāo)分子。本論文主要包括以下內(nèi)容:1、探針T1和探針T2是以香豆素做為熒光團(tuán),用來(lái)檢測(cè)亞硫酸根的熒光探針。探針T1和T2分別在CH3CN-HEPES(0.02 M,pH 7.4)(V/V=2:8)的緩沖溶液,CH3CN-PBS(0.02 M,pH 7.4)(V/V=2:8)的緩沖溶液中進(jìn)行測(cè)試。探針T1對(duì)亞硫酸根表現(xiàn)出較好的選擇性,探針T1還被成功的應(yīng)用到Hela細(xì)胞中對(duì)亞硫酸根檢測(cè)。2、探針T3通過(guò)半菁染料和N-乙基-3,6-二甲醛基咔唑縮合得到。在光物理性質(zhì)的研究中,我們觀(guān)察到探針T3在DMF-PBS(0.01 M,pH=7.4)(V:V=3:7)緩沖液中加入HSO3-后,溶液的顏色發(fā)生了從淡黃色變成了無(wú)色的變化,并伴隨著熒光增強(qiáng)的響應(yīng)。探針T3被成功的應(yīng)用到Hela細(xì)胞和秀麗線(xiàn)蟲(chóng)當(dāng)中,通過(guò)熒光強(qiáng)度的改變定量的測(cè)定外源性HSO3-的含量。3、探針T4以探針T3做為基礎(chǔ),通過(guò)T3中的醛與鹽酸羥氨作用生成肟,用來(lái)檢測(cè)次氯酸根。T4在DMF-PBS(0.01 M,pH 7.4)(V/V=4:6)的緩沖溶液中測(cè)試,加入次氯酸根后,出現(xiàn)熒光增強(qiáng)的響應(yīng),在各種干擾離子的存在下,探針T4表現(xiàn)出優(yōu)異的選擇性。探針T4被成功地應(yīng)用到生物熒光成像檢測(cè)Hela細(xì)胞和秀麗線(xiàn)蟲(chóng)中的次氯酸跟離子,通過(guò)熒光強(qiáng)度的變化可以測(cè)定外源性ClO-的含量。
[Abstract]:Sulfur dioxide derivatives are often used as food preservatives in daily life to prevent food oxidation and bacterial growth. As industrialization progressed, the use of coal, oil and fossil fuels increased, causing SO2 to be pumped into the air. The low concentration of sulfur dioxide derivatives has little effect on human body. However, when the concentration of sulfur dioxide derivatives exceeds a certain range, it will cause certain harm to human health, such as respiratory diseases, cancer and so on. Hypochloric acid is a by-product produced by human body during natural oxidation and metabolism. Hypochlorite is closely related to various physiological and pathological processes of organism. When the concentration of hypochloric acid in organisms is too high, it can cause cancer, arthritis and other diseases. Therefore, quantitative detection of sulfur dioxide derivatives and hypochlorite by fluorescence probe technique is of great significance. In this paper, fluorescent probes were designed and synthesized for the detection of T1 and T2H _ 2, T3) and T4), and their photophysical properties and biological applications were studied. It is found that these probes have excellent selectivity and good spectral signal response. These probes have been successfully used to detect target molecules in living cells and nematodes. The main contents of this thesis are as follows: probe T1 and probe T2 are fluorescent probes using coumarin as fluorescence group to detect sulfite radical. The probe T1 and T2 were tested in the buffer solution CH3CN-PBSN 0.02 MN pH 7.4 / V / V 2: 8, respectively, in the buffer solution Ch _ 3CN-PBS _ 2: 8 (pH 7.4 / V / V _ (2: 8)). Probe T1 showed good selectivity for sulfite, and probe T1 was also successfully applied to detect sulfite in Hela cells. Probe T3 was obtained by condensation of hemicyanine dye with N-ethyl-3-dimethylcarbazole. In the study of photophysical properties, we observed that the color of the solution changed from light yellow to colorless after the addition of HSO _ 3- in the buffer solution of DMF-PBS(0.01 M _ 3 pH = 7.4 / V: v _ (3: 7), which was accompanied by the response of fluorescence enhancement. Probe T3 was successfully applied to Hela cells and nematodes. The content of exogenous HSO3- was determined quantitatively by changing the fluorescence intensity. The probe T4 was based on probe T3 and formed oxime through the reaction of aldehyde in T3 with hydroxylamine hydrochloride. The method was used to detect hypochlorite in DMF-PBS(0.01 pH 7.4 V / V 4: 6 buffer solution. After adding hypochlorite, the fluorescence response was enhanced. In the presence of various interference ions, the probe T4 showed excellent selectivity. The probe T4 was successfully applied to the detection of hypochloric acid and ions in Hela cells and nematode, and the content of exogenous ClO- could be determined by the change of fluorescence intensity.
