石墨爐原子吸收法測定大鼠飲用富硒綠茶后硒元素在大鼠組織中的含量

【摘要】:通過建立測定大鼠血漿、心臟、肝臟、睪丸、胰腺、肺、指甲、腦、腎臟和肌肉組織中硒含量的石墨爐原子吸收法,測定大鼠飲用富硒綠茶后硒元素在各組織中的含量和分布情況。將大鼠隨機分為富硒綠茶組(10 g/kg)、非富硒綠茶組(10 g/kg)和空白組,每組6只,連續(xù)灌胃30 d,每天一次。采用濕法消化法處理大鼠組織樣品,以石墨爐原子吸收法測定其硒的含量。采用標(biāo)準(zhǔn)加入法制備工作曲線,經(jīng)方法學(xué)驗證,各組織硒的含量在10~100 ng/mL范圍內(nèi)線性關(guān)系良好(r≥0.99136),批間和批內(nèi)精密度小于12.2%,平均準(zhǔn)確度89.5%~106.9%之間,平均回收率86.1%~95.1%之間;組織分布研究表明富硒綠茶組與空白組相比,其心臟、睪丸、肺、肝臟、腎臟和胰腺的硒含量有明顯增加;與非富硒綠茶組比,其胰腺的硒含量有明顯增加。本研究通過優(yōu)化組織樣品預(yù)處理過程和分別為各組織制備工作曲線所建立的石墨爐原子吸收法準(zhǔn)確度好,重復(fù)性高,可以適用于測定大鼠飲用富硒綠茶后各組織中硒的含量,大鼠飲用富硒綠茶后能夠吸收其硒元素,各組織中的硒含量在安全范圍之內(nèi)。
[Abstract]:A graphite furnace atomic absorption spectrometry was established for the determination of selenium in plasma, heart, liver, testis, pancreas, lung, fingernail, brain, kidney and muscle of rats.The content and distribution of selenium in different tissues of rats after drinking selenium-rich green tea were determined.The rats were randomly divided into two groups: se enriched green tea group (10 g / kg), non se rich green tea group (10 g / kg) and blank group (6 rats in each group).The content of selenium in rat tissue was determined by graphite furnace atomic absorption spectrometry (GFAAS).The standard addition method was used to prepare the working curve.
