基于化學(xué)衍生化的靶向脂質(zhì)組學(xué)質(zhì)譜分析研究進(jìn)展

【摘要】:脂質(zhì)組學(xué)研究與一些重大疾病(如心血管病、糖尿病、腫瘤等)密切相關(guān),是當(dāng)今生命科學(xué)研究的重要領(lǐng)域.質(zhì)譜分析技術(shù)是目前脂質(zhì)組學(xué)研究的核心手段之一.化學(xué)衍生化策略可顯著提高基于鳥槍法或液相色譜-質(zhì)譜聯(lián)用方法的脂質(zhì)組學(xué)分析性能,近年來已有一些相關(guān)研究報(bào)道.本文重點(diǎn)介紹了基于化學(xué)衍生化的質(zhì)譜分析方法在靶向脂質(zhì)組學(xué)研究中的進(jìn)展,并提出了問題和展望.希望在衍生化試劑的設(shè)計(jì)與合成,特別是具有顯著優(yōu)勢(shì)的穩(wěn)定同位素標(biāo)記衍生試劑開發(fā),以及新的衍生化策略和相應(yīng)的質(zhì)譜分析方法的開發(fā)與應(yīng)用方面起到一定的推動(dòng)作用.
[Abstract]:Lipid studies are closely associated with major diseases such as cardiovascular disease, diabetes, cancer, etc. Mass spectrometry (MS) is one of the core methods of liposome research. The chemical derivatization strategy can significantly improve the performance of liposome analysis based on shotgun method or liquid chromatography-mass spectrometry (LC-MS). In recent years, some related studies have been reported. This paper focuses on the development of mass spectrometry based on chemical derivation in the study of targeted liposome, and puts forward some problems and prospects. It is hoped that the design and synthesis of derivatization reagents will be carried out. In particular, the development of stable isotope labeled derivatives, as well as the development and application of new derivatization strategies and the corresponding mass spectrometric analysis methods, have played a certain role in promoting the development and application of stable isotope labeled derivatives.
