【摘要】:本論文研究了質(zhì)子酸催化的烯丙基亞胺離子分子內(nèi)環(huán)化反應(yīng),主要包括以下三章內(nèi)容:第一章:簡(jiǎn)述了(1)質(zhì)子酸催化劑催化反應(yīng)的歷程(2)質(zhì)子酸催化劑的發(fā)展前景(3)亞胺離子中間體在環(huán)化反應(yīng)中的應(yīng)用。第二章:三氟乙酸催化的烯丙基亞胺離子環(huán)化構(gòu)建環(huán)戊并[b]吲哚骨架反應(yīng)研究。環(huán)戊并[b]吲哚骨架廣泛存在于天然產(chǎn)物和藥物分子中,含有環(huán)戊并[b]吲哚骨架結(jié)構(gòu)的化合物具有抗腫瘤、抗菌、殺蟲等功效,由此可見,發(fā)展高效構(gòu)建環(huán)戊并[b]吲哚骨架的方法具有重要意義。本課題發(fā)展的三氟乙酸催化吲哚取代烯丙基亞胺離子分子內(nèi)環(huán)化反應(yīng)構(gòu)建吲哚衍生物目前尚未見其它報(bào)道。反應(yīng)過程中,由于催化劑TFA具有吸濕發(fā)煙、腐蝕性,所以采用硅膠負(fù)載;而且生成水為唯一副產(chǎn)物。該方法不僅豐富了質(zhì)子酸催化劑在催化合成中的應(yīng)用,而且為吲哚衍生物的合成提供了研究基礎(chǔ)。第三章:PPA催化的烯丙基亞胺離子環(huán)化構(gòu)建茚類衍生物反應(yīng)研究。茚環(huán)骨架是許多天然產(chǎn)物的核心骨架,廣泛存在于生物活性分子、功能材料和過渡金屬絡(luò)合物中,其中金屬-茚基配合物在催化烯烴聚合中具有顯著效果,因此,合成茚類衍生物具有重要意義。多磷酸(PPA)是一種強(qiáng)無機(jī)酸,具有較強(qiáng)的脫水性能,廣泛用于分子內(nèi)和分子間;,雜環(huán)合成和酸催化反應(yīng)中。本課題提供了一種獨(dú)特的、新穎的PPA催化烯丙基亞胺離子中間體采用外型關(guān)環(huán)模式構(gòu)建茚環(huán)骨架的方法。該反應(yīng)具有以下優(yōu)點(diǎn):原料便宜易得、底物普適性好(不僅適用于富電子體系,對(duì)于缺電子醛也很有效)、無金屬參與、原子經(jīng)濟(jì)性高。本課題為合成茚烷衍生物提供了良好的研究基礎(chǔ)。
[Abstract]:In this paper, the intramolecular cyclization of allyl imines catalyzed by proton acid was studied. The main contents are as follows: chapter 1: the course of catalytic reaction of proton acid catalyst 2) the prospect of proton acid catalyst) the application of imine ion intermediate in cyclization reaction. Chapter 2: trifluoroacetic acid. Cyclization of Allyl imines to Cyclopentazo [b] Indole cytoskeleton. Cyclopentazo [b] indole cytoskeleton is widely present in natural products and drug molecules. Compounds containing cyclopentyl [b] indole skeleton have anti-tumor, antibacterial and insecticidal properties. It is of great significance to develop an efficient method for the construction of cyclization of cyclization of cyclization of cyclization of cyclization of cyclization of indole-substituted allyl imide ion with trifluoroacetic acid. Because the catalyst TFA is hygroscopically smoky and corrosive, it is supported by silica gel and water is the only by-product. This method not only enriches the application of proton acid catalyst in catalytic synthesis. It also provides a basis for the synthesis of indole derivatives. Chapter 3: PPA catalyzed cyclization of allyl imines to indene derivatives. Indenyl cytoskeleton is the core skeleton of many natural products and widely exists in bioactive molecules. In functional materials and transition metal complexes, the metal-indenyl complexes have remarkable effect in catalytic olefin polymerization, so the synthesis of indene derivatives is of great significance. It is widely used in intramolecular and intermolecular acylation, heterocyclic synthesis and acid-catalyzed reactions. A novel PPA catalytic method for the construction of indene cytoskeleton in allyl imide ion intermediates using external ring closure mode is presented. The reaction has the following advantages: the raw material is cheap and easy to obtain, the substrate is universal (not only suitable for electron-rich systems, but also suitable for electronic systems). For the lack of electron aldehydes is also very effective, no metal participation, high atomic economy. This topic provides a good basis for the synthesis of ninhydrin derivatives.
