溴代乙酰葡萄糖和溴代乙酰鼠李糖的便捷合成

【摘要】:以高氯酸為催化劑,D-葡萄糖和L-鼠李糖分別與乙酸酐反應(yīng),制得相應(yīng)的全乙;,不經(jīng)分離直接加入乙酰溴和甲醇,利用反應(yīng)生成的溴化氫原位發(fā)生溴代反應(yīng),一鍋法制備了溴代乙酰葡萄糖和溴代乙酰鼠李糖,收率分別達(dá)到92.5%和85.6%。從產(chǎn)品結(jié)晶后的母液中,以干燥的二氯甲烷作溶劑,采用重結(jié)晶法分離得到溴代乙酰鼠李糖合成的副產(chǎn)物2,3,4-三-O-乙;罄钐。產(chǎn)品和副產(chǎn)物的結(jié)構(gòu)均經(jīng)過核磁共振氫譜、碳譜和高分辨質(zhì)譜確認(rèn)。
[Abstract]:Using perchloric acid as catalyst, D-glucose and L-rhamnose were reacted with acetic anhydride respectively to produce corresponding total acetylated sugar. Acetyl bromide and methanol were added directly without separation. Bromoacetyl glucose and bromoacetyl rhamnose were prepared by one-pot method in 92.5% and 85.6% yields, respectively. Dry dichloromethane was used as solvent from the crystallized mother liquor of the product. The by-product of bromoacetyl rhamnose was isolated by recrystallization method. The structure of the by-product was confirmed by NMR, carbon spectrum and high resolution mass spectrometry.
