【摘要】：研究頭孢噻肟(cefotaxime,CTX)在雞血漿中代謝規(guī)律,建立同時檢測血漿中CTX及其主要代謝物去乙酰頭孢噻肟(desacetylcefotaxime,DCTX)的超高效液相色譜-串聯(lián)質(zhì)譜方法,以10 mg/kg的體質(zhì)量劑量給藥實驗蛋雞,并對給藥后不同時間點采集的血液樣品,進行CTX及其主要代謝物DCTX含量的檢測。結(jié)果表明,所建方法中CTX和DCTX的檢出限分別為0.07、0.14μg/L;定量限分別為0.23、0.99μg/L�；厥章史謩e為86.7%~95.2%、89.5%~112%,相對標準偏差不大于7.7%,滿足殘留分析方法的要求;CTX在蛋雞體內(nèi)分布迅速,給藥后即刻達到峰值(56.34 mg/L),且代謝較快,給藥24 h后檢測值低于3.0μg/L,而主要代謝物DCTX在給藥45 min后達到峰值(10.22 mg/L),消除相對緩慢,給藥24 h時檢測值仍可達到54.0μg/L。
[Abstract]:To study the metabolic characteristics of cefotaxime (CTX) in chicken plasma, and to establish a high performance liquid chromatography-tandem mass spectrometry method for simultaneous determination of CTX and its main metabolite, desacetylcefotaxime, in chicken plasma. The contents of CTX and its main metabolite DCTX in blood samples collected at different time points after administration were determined. The detection limits of CTX and DCTX were 0.07 ~ 0.14 渭 g / L and 0.23 ~ 0.99 渭 g / L, respectively. The recoveries of CTX and DCTX were 86.7% and 95.2 渭 g / L, respectively. The relative standard deviation was not more than 7.70.CTX distributed rapidly in layers. The peak value (56.34 mg / L) was reached immediately after administration, and the metabolic rate was faster. The detection value of DCTX was lower than 3.0 渭 g / L 24 h after administration, while the peak value of DCTX reached 10.22 mg / L after 45 min administration, and the elimination was relatively slow. The detection value was still up to 54.0 渭 g / L at 24 h.
