二醇基硅膠固相萃取-多維質譜對烏鱧組織脂質組學分析

【摘要】:建立以二醇基鍵合硅膠(Si O2/diol)為填料的固相萃取(solid-phase extraction,SPE)和基于多維度串聯(lián)質譜的鳥槍法分離純化并測定烏鱧肌肉組織中的磷脂。利用Folch法和SPE法提取并純化磷脂,比較3種不同填料SPE柱的萃取效果,優(yōu)化SPE參數(shù),最佳條件:上樣量100?μL,淋洗液p H值為2,洗脫液為體積分數(shù)50%乙腈溶液1 m L。所得提取物以直接進樣的方法流動注射入電噴霧離子源,經(jīng)三重四極桿質譜的母離子掃描和中性丟失掃描模式,實現(xiàn)對磷脂分子種類的鑒定和定量。同時,通過多維度串聯(lián)質譜分析,得到各個磷脂分子中2條脂肪酸鏈長度和不飽和度信息。實驗結果顯示,烏鱧樣品中成功檢出磷脂酰膽堿、磷脂酰乙醇胺、磷脂酰肌醇和磷脂酰絲氨酸4類磷脂,共計67種磷脂分子,且大部分磷脂均含有不飽和脂肪酸鏈。本方法簡單高效,分析準確快捷,結果穩(wěn)定。
[Abstract]:Solid phase extraction (SPE) with diol bonded silica gel (Sio _ 2 / diol) as filler and bird gun method based on multi-dimensional tandem mass spectrometry were used to isolate and determine phospholipid in the muscle tissue of snakehead Argus Argus. Phospholipid was extracted and purified by Folch and SPE. The extraction efficiency of three different packing SPE columns was compared and the SPE parameters were optimized. The optimum conditions were as follows: sample amount 100? 渭 L, pH value of eluant was 2, elution solution was 50% acetonitrile solution 1 mL by volume fraction. The extract was injected into electrospray ion source by direct injection method, and was scanned by mother ion and neutral loss by triple quadrupole mass spectrometry. The identification and quantification of phospholipids were realized. At the same time, the length and unsaturation information of two fatty acid chains in each phospholipid molecule were obtained by multi-dimensional tandem mass spectrometry. Phosphatidylcholine, phosphatidylethanolamine, phosphatidylinositol and phosphatidyl serine were successfully detected in snakehead samples. The analysis is accurate and quick, and the result is stable.
