芳香族酰腙及多吡啶羧酸類有機(jī)錫配合物的合成、結(jié)構(gòu)及性質(zhì)研究

【摘要】:有機(jī)錫配合物以其復(fù)雜的結(jié)構(gòu)、優(yōu)異的性能而越來(lái)越受到科學(xué)家們的青睞。尤其是自二十世紀(jì)八十年代以來(lái),人們發(fā)現(xiàn)有機(jī)錫配合物具有良好的生物活性,因此該類配合物已成為醫(yī)學(xué)、藥學(xué)、生物學(xué)及化學(xué)等諸多學(xué)科的熱門研究對(duì)象。由于芳香族酰腙類以及多吡啶羧酸類Schiff堿化合物其本身就具有的一定的抑菌、抗癌等生物活性以及靈活多樣的配位形式,所以,本文以芳香酰肼和取代水楊醛為原料設(shè)計(jì)合成了芳香族酰腙類配體,同時(shí)又以鄰啡咯啉為原料設(shè)計(jì)合成了多吡啶羧酸類配體。然后用合成的配體與有機(jī)錫化合物進(jìn)行反應(yīng),得到了一系列新型有機(jī)錫配合物,并通過(guò)元素分析、核磁共振、紅外光譜以及X-射線單晶衍射等方法對(duì)其結(jié)構(gòu)進(jìn)行了表征。并對(duì)配合物的體外抗癌活性和對(duì)細(xì)胞內(nèi)活性氧的誘導(dǎo)活性以及與小牛胸腺DNA(CT-DNA)和牛血清白蛋白(BSA)的結(jié)合作用進(jìn)行了研究。以下是主要工作:1.設(shè)計(jì)合成了四種酰腙類Schiff堿配體和一種多吡啶羧酸類Schiff堿配體:2-羥基-3-甲氧基苯甲醛水楊酰腙、5-氯水楊醛-4-吡啶甲酰腙、5-氯水楊醛水楊酰腙、2-羥基-4-甲氧基苯甲醛-4-吡啶甲酰腙、對(duì)甲;郊姿峥s-4,5-二氮芴-9-腙。同時(shí)利用X-射線單晶衍射儀對(duì)其中的兩個(gè)配體進(jìn)行了結(jié)構(gòu)表征。2.設(shè)計(jì)合成了2-羥基-3-甲氧基苯甲醛水楊酰腙類有機(jī)錫配合物,并得到了五種有機(jī)錫配合物的單晶結(jié)構(gòu)。該系列酰腙類有機(jī)錫配合物多以單核和二聚體形式存在。3.設(shè)計(jì)合成了5-氯水楊醛-4-吡啶甲酰腙有機(jī)錫配合物和5-氯水楊醛水楊酰腙有機(jī)錫配合物,得到了三種有機(jī)錫配合物的單晶結(jié)構(gòu)。結(jié)果表明,三種配合物結(jié)構(gòu)類似,均為三核結(jié)構(gòu)。分子內(nèi)錫原子成中心對(duì)稱,中心錫原子為六配位的畸形八面體結(jié)構(gòu),兩端的錫原子,形成五配位扭曲的三角雙錐構(gòu)型。4.合成了2-羥基-4-甲氧基苯甲醛-4-吡啶甲酰腙類有機(jī)錫配合物,得到了兩種有機(jī)錫配合物的單晶結(jié)構(gòu)。結(jié)構(gòu)特征與2-羥基-3-甲氧基苯甲醛水楊酰腙類有機(jī)錫配合物相似。5.合成了對(duì)甲;郊姿峥s-4,5-二氮芴-9-腙有機(jī)錫配合物,得到了一種配合物的晶體,并且還意外得到了一個(gè)金屬鎂的配合物。該有機(jī)錫配合物是四核的中心對(duì)稱結(jié)構(gòu),每個(gè)錫原子都采用六配位扭曲的八面體構(gòu)型,分子的平面性較好,所有非氫原子基本上都在同一個(gè)平面內(nèi)。6.性質(zhì)研究通過(guò)MTT法對(duì)合成的配體和有機(jī)錫配合物進(jìn)行了體外抗癌活性檢測(cè),并研究了部分配合物對(duì)細(xì)胞內(nèi)活性氧的誘導(dǎo)活性。另外,還運(yùn)用紫外、熒光以及圓二色等光譜分析方法,對(duì)酰腙類有機(jī)錫配合物與CT-DNA和BSA的相互作用進(jìn)行了研究。結(jié)果表明,多數(shù)酰腙類有機(jī)錫配合物具有良好的體外抗腫瘤活性,并與DNA和BSA具有較強(qiáng)的相互作用,有機(jī)錫配合物都可以使BSA發(fā)生靜態(tài)熒光猝滅現(xiàn)象。
[Abstract]:Organotin Complexes with the complex structure, excellent performance, more and more scientists of all ages. Especially since 1980s, people found that the organotin compounds have good biological activity, so the complex has become a hot research object of medicine, pharmacy, biology and chemistry and many other disciplines. Since aromatic hydrazone and pyridine carboxylic acid Schiff base compounds itself has certain antibacterial, anticancer activity and flexible coordination form, therefore, the aromatic hydrazone ligand have been synthesized with aromatic hydrazide and substituted salicylaldehyde, and adjacent to the coffee phenanthroline pyridine carboxylic acid ligand is synthesized. Then design synthetic ligands to react with organic tin compounds, a series of novel organotin complexes, and characterized by elemental analysis, nuclear Magnetic resonance, infrared spectrum and X- ray diffraction were used to characterize its structure. Inducing activity of in vitro anticancer activity of the complexes and the intracellular reactive oxygen species and calf thymus DNA (CT-DNA) and bovine serum albumin (BSA) binding were studied. In addition, using ultraviolet fluorescence method, and round two color spectrum analysis of acyl hydrazone Organotin Complexes with CT-DNA interaction and BSA were studied. The results showed that the majority of hydrazone organotin complexes have good antitumor activity in vitro, and has strong interaction with DNA and BSA, organotin complexes can make BSA The phenomenon of static fluorescence quenching.
