超聲萃取高效液相色譜法測(cè)定諾麗果粉中的莨菪亭、蘆丁和槲皮素含量

【摘要】:采用超聲萃取高效液相色譜法測(cè)定諾麗果粉中的莨菪亭、蘆丁和槲皮素含量。為了提高萃取效率,實(shí)驗(yàn)系統(tǒng)地優(yōu)化了影響萃取效率的因素(有機(jī)溶劑種類、有機(jī)溶劑與水的比例、萃取時(shí)間、萃取溫度和超聲頻率)。結(jié)果表明,最佳萃取條件:萃取劑為體積分?jǐn)?shù)80%甲醇溶液,萃取時(shí)間為30 min,萃取溫度為20℃,超聲頻率為45 k Hz。采用依利特Hypersil ODS柱(4.6 mm×250 mm,5μm),以甲醇-0.1%三氟乙酸-乙腈溶液作為流動(dòng)相進(jìn)行梯度淋洗,流速1.0 m L/min,在檢測(cè)波長365 nm,柱溫25℃條件下,對(duì)莨菪亭、蘆丁和槲皮素分離檢測(cè)。方法具有良好的線性關(guān)系(r2≥0.999 8)、檢出限(≤1.30μg/L)、回收率(90.67%~113.00%)和重復(fù)性(相對(duì)標(biāo)準(zhǔn)偏差不大于1.46%,n=8)。該方法準(zhǔn)確可靠,可用于定性、定量測(cè)定諾麗果粉樣品中的莨菪亭、蘆丁和槲皮素。
[Abstract]:The contents of scopoletin, rutin and quercetin in Nori fruit powder were determined by ultrasonic extraction high performance liquid chromatography. In order to improve the extraction efficiency, the factors affecting the extraction efficiency were systematically optimized. The ratio of organic solvent to water, extraction time, extraction temperature and ultrasonic frequency are determined.
