溶酶體熒光探針研究新進展

【摘要】:溶酶體在細胞各種生命活動,如物質代謝、細胞膜循環(huán)、細胞凋亡中發(fā)揮著重要作用.將溶酶體可視化,并對其活性物種、特定微環(huán)境及關鍵生理過程進行檢測,不僅有助于理解溶酶體參與生命活動的分子機制,而且對疾病的治療具有重要的指導意義.近年來,許多靶向溶酶體的熒光探針被相繼報道,其檢測對象包括還原性物種、活性氧、活性氮、氫離子、金屬離子、陰離子以及生物酶等,還包括對溫度、粘度、極性、p H等微環(huán)境的檢測.溶酶體的熒光探針的定位機制分為三類:利用溶酶體的酸性生理環(huán)境、利用溶酶體的物質代謝功能以及利用溶酶體特殊的膜蛋白或水解酶.對近年來出現的主要溶酶體熒光探針進行了總結與評述,展望了溶酶體熒光探針應用于探究重要細胞生理過程的分子機制,以及相關疾病診斷方面的發(fā)展趨勢.
[Abstract]:Lysosomes play an important role in various cell life activities, such as substance metabolism, cell membrane circulation and apoptosis. The lysosomes are visualized and detected for their active species, specific microenvironments and key physiological processes. Not only is it helpful to understand the molecular mechanism of lysosome's involvement in life, but it is also of great significance in the treatment of diseases. In recent years, a number of fluorescent probes targeting lysosomes have been reported, including the detection of reductive species. Reactive oxygen species, active nitrogen, hydrogen ions, metal ions, anions, biological enzymes, and so on, including temperature, viscosity, The localization mechanism of lysosomal fluorescence probe is divided into three categories: using the acidic physiological environment of lysosome, Based on the metabolic function of lysosome and the special membrane protein or hydrolase of lysosome, the main lysosomal fluorescent probes in recent years were summarized and reviewed. The application of lysosomal fluorescence probe to the molecular mechanism of important cellular physiological processes and the development trend of diagnosis of related diseases are prospected.
