pH敏感納米凝膠的制備及其在siRNA轉(zhuǎn)染中的應(yīng)用

【摘要】:通過(guò)分散聚合的方法,以改性了雙鍵的葡聚糖(Dex-AA)作為交聯(lián)劑,甲基丙烯酸二甲氨基乙酯(DMAEMA)作為單體,過(guò)硫酸銨(APS)和四甲基乙二胺(TEMED)分別作為引發(fā)劑和助引發(fā)劑,合成了不同交聯(lián)度的、具有pH敏感內(nèi)吞增強(qiáng)作用的葡聚糖納米凝膠(DD-NGs),并測(cè)試了其復(fù)合siRNA進(jìn)行轉(zhuǎn)染的能力.實(shí)驗(yàn)結(jié)果表明,該納米凝膠表面帶有正電荷,具有較好的擔(dān)載siRNA進(jìn)入腫瘤細(xì)胞并沉默基因的能力,且具有pH響應(yīng)粒徑變化的性質(zhì).在pH=7.4的體液環(huán)境中,納米凝膠與基因的復(fù)合物粒子較小;在腫瘤酸性(pH=6.8)條件下,納米凝膠與基因的復(fù)合物粒子變大,顯著地增強(qiáng)了腫瘤細(xì)胞對(duì)納米凝膠與基因復(fù)合物的內(nèi)吞.
[Abstract]:The modified dextran (Dex-AA) was used as crosslinking agent and dimethylaminoethyl methacrylate (DMAEMA) was used as monomer by dispersion polymerization. Ammonium persulfate (APS) and tetramethyl ethylenediamine (TEMED) were used as initiators and co-initiators, respectively. The pH sensitive endocytosis enhanced dextran nanogel DD-NGsN was tested and its ability to transfect the composite siRNA was tested. The experimental results showed that the surface of the nanogel was positively charged. It has the ability of carrying siRNA into tumor cells and silencing genes, and has the property of pH response to particle size change. In the humoral environment of pH=7.4. The composite particles of nanogel and gene were smaller. Under the condition of tumor acidic pH = 6.8), the complex particles of nano-gel and gene became larger, which significantly enhanced the endocytosis of tumor cells to nano-gel and gene complex.
